黃原膠對前交叉韌帶切斷術(shù)所致骨關(guān)節(jié)炎的療效及其機制

【摘要】:本研究評價黃原膠(xanthan gum,XG)對前交叉韌帶切斷術(shù)(anterior cruciate ligament transaction,ACLT)所致的兔膝關(guān)節(jié)骨關(guān)節(jié)炎(osteoarthritis,OA)模型中軟骨損傷的保護作用,并探討XG對OA軟骨中caspase-3、Bax蛋白表達的影響。將60只雄性新西蘭大耳白兔按隨機數(shù)字表法分為6組,每組10只,隨機選取1組為正常對照組(control組),剩余5組右膝采用ACLT建立OA模型,并根據(jù)藥物干預不同分為模型組(model組)、XG-0.6 mg·k~(g-1)、XG-1.2 mg·k~(g-1)、XG-2.4 mg·k~(g-1)治療組和玻璃酸鈉(sodium hyaluronate,SH-1.2 mg·k~(g-1))治療組。測量治療過程中各組兔膝關(guān)節(jié)局部溫度和膝關(guān)節(jié)寬度;治療結(jié)束后,對各組兔關(guān)節(jié)進行大體形態(tài)學觀察;通過HE染色對各組兔關(guān)節(jié)軟骨組織病理學形態(tài)進行觀察;Western blot法檢測各組家兔軟骨細胞中Bax和caspase-3的激活態(tài)cleaved caspase-3含量。實驗結(jié)果表明,XG可抑制OA所致的膝關(guān)節(jié)局部溫度的上升和膝關(guān)節(jié)寬度的增加,且具有劑量依賴性;XG具有改善由OA所致的股骨髁和脛骨平臺形態(tài)學異常和組織損傷的作用;Western blot結(jié)果表明:與control組相比,model組和XG-0.6 mg·k~(g-1)組家兔膝關(guān)節(jié)軟骨細胞中Bax和cleaved caspase-3水平顯著增加(P0.01),model組和XG-0.6 mg·k~(g-1)組間兔膝關(guān)節(jié)軟骨細胞中Bax和cleaved caspase-3水平無顯著性差別(P0.05),且明顯高于XG-1.2 mg·k~(g-1)和XG-2.4 mg·k~(g-1)組(P0.01),XG-2.4 mg·k~(g-1)和XG-1.2 mg·k~(g-1)組間兔骨關(guān)節(jié)軟骨細胞中cleaved caspase-3水平無顯著性差別(P0.05),XG-2.4 mg·k~(g-1)組兔骨關(guān)節(jié)軟骨細胞中Bax水平低于XG-1.2 mg·k~(g-1)組(P0.05)。綜上所述,XG能夠有效地保護OA中的軟骨損傷,并能夠抑制OA軟骨中Bax和caspase-3蛋白的表達。
[Abstract]:The purpose of this study was to evaluate the protective effect of xanthan gumma XG on cartilage injury induced by anterior cruciate ligament transaction (ACLT) in rabbits with osteoarthritis of knee joint, and to investigate the effect of XG on the expression of caspase-3Bax protein in OA cartilage.Sixty male New Zealand white rabbits were randomly divided into 6 groups with 10 rabbits in each group. One group was randomly selected as control group, and the remaining 5 groups were treated with ACLT to establish OA model.According to the drug intervention, the model group was divided into two groups: model group (XG-0.6 mg / kg) and sodium hyaluronate sodium hyaluronateSH-1.2 mg / kg ~ (-1) (treated with XG-1.2 mg / kg ~ (-1) + XG-2.4mg / kg ~ (-1)) respectively, and the model group was treated with sodium hyaluronate SH-1.2 mg / kg ~ (-1) sodium hyaluronate (SH-1.2 mg / kg ~ (-1)).The local temperature and width of knee joint in each group were measured during the treatment, and the gross morphology of each group was observed after the treatment.The histopathological morphology of articular cartilage in each group was observed by HE staining. The contents of Bax and caspase-3 active cleaved caspase-3 in chondrocytes of each group were detected by Western blot method.The experimental results showed that XG could inhibit the rise of knee joint temperature and the increase of knee joint width induced by OA.The results of Western blot showed that compared with control group and XG-0.6 mg / kg -1 group, Bax and cleaved in articular chondrocytes of rabbit knee joint were improved.There was no significant difference in the levels of Bax and cleaved caspase-3 in rabbit knee joint chondrocytes between the two groups (P 0.05) and XG-2.4 mg / kg ~ (1) and XG-2.4 (mg / kg ~ (-1)) respectively. There was no significant difference in cleaved caspase-3 level in articular chondrocytes between the two groups (P _ (0.01) and XG-0.6 mg / kg ~ (-1)) and XG-1.2 (mg / kg ~ (1)) groups. (P _ (0.05)) and XG-2.4 (mg / kg ~ (-1)), respectively, there was no significant difference in cleaved caspase-3 level in articular chondrocytes between the two groups (P _ (0.01) and XG-1.2 mg / kg ~ (-1)).The level of Bax in rabbit osteoarticular chondrocytes was lower than that in XG-1.2 group (P 0.05).In conclusion, XG can effectively protect cartilage injury and inhibit the expression of Bax and caspase-3 in OA cartilage.
