【摘要】：該研究旨在建立一種新型的載多烯紫杉醇的TPGS修飾前體囊泡口服遞藥系統(tǒng)(Doce-TPGS-PN),并對(duì)其水化后的物理化學(xué)特性和體外釋放進(jìn)行考察。首先,以制得前囊泡水化后的粒徑、包封率和突釋量為指標(biāo),考察脂質(zhì)材料的種類、TPGS修飾方法、TPGS加入量及表面活性劑和膽固醇的比例對(duì)前囊泡水化后性質(zhì)的影響,篩選最優(yōu)處方和制備工藝。其次,對(duì)最優(yōu)處方制得DTX-TPGS-PN的形態(tài)、粒徑、表面電荷、包封率及體外釋放行為進(jìn)行考察。結(jié)果表明,Doce-TPGS-PN系統(tǒng)水化后,粒徑為(93±6.5)nm,Zeta電位為(-83.95±3.69)m V,包封率(97.31±0.60)%,在PBS中的釋放分為前0.5 h的速釋相和之后的緩釋相。實(shí)驗(yàn)證明,該研究成功構(gòu)建了Doce-TPGS-PN這一新型口服給藥系統(tǒng),且其具有良好性質(zhì)。
[Abstract]:The aim of this study was to establish a novel doce-TPGS-PNN system with paclitaxel loaded TPGS modified precursor vesicles, and to investigate its physicochemical properties and in vitro release after hydration.Firstly, the particle size, entrapment efficiency and sudden release of the prepared anterior vesicle after hydration were used as indexes to investigate the effects of the type of lipid materials modified by TPGS and the ratio of surfactant to cholesterol on the properties of the vesicle after hydration.To select the best prescription and preparation technology.Secondly, the morphology, particle size, surface charge, encapsulation efficiency and in vitro release behavior of DTX-TPGS-PN were investigated.The results showed that after hydration of Doce-TPGS-PN system, the particle size was 93 鹵6.5nmGS-PN, the Zeta potential was -83.95 鹵3.69mV, the entrapment efficiency was 97.31 鹵0.60mV, and the release in PBS was divided into two phases: the first 0.5 h rapid release phase and the later slow release phase.The experimental results show that Doce-TPGS-PN, a new oral drug delivery system, is successfully constructed and has good properties.
