【摘要】：共價(jià)藥物是一類能與靶標(biāo)形成共價(jià)鍵從而發(fā)揮生物學(xué)功能的藥物。隨著多個(gè)激酶抑制劑類抗腫瘤共價(jià)藥物的上市,共價(jià)藥物重新回到藥物發(fā)現(xiàn)的視野并成為抗腫瘤化療藥物的研究熱點(diǎn)。本文從共價(jià)藥物的作用機(jī)制、藥理學(xué)優(yōu)勢(shì)、開(kāi)發(fā)策略以及近年來(lái)出現(xiàn)的抗腫瘤共價(jià)藥物及候選藥物的研究進(jìn)展進(jìn)行綜述,為新型抗腫瘤共價(jià)藥物的設(shè)計(jì)提供參考。
[Abstract]:Covalent drugs are a class of drugs that can form covalent bonds with the target to play a biological role.With the coming into market of many kinase inhibitors, covalent drugs return to the field of drug discovery and become the research hotspot of antitumor chemotherapeutic drugs.In this paper, the mechanism of covalent drugs, pharmacological advantages, development strategies and research progress of covalent drugs and candidate drugs in recent years are reviewed in order to provide references for the design of new antitumor covalent drugs.
