瑞舒伐他汀對同型半胱氨酸損傷人臍靜脈內皮細胞血管細胞間黏附分子-1表達的影響

【摘要】:目的探討瑞舒伐他汀(Rosu)對同型半胱氨酸(Hcy)損傷人臍靜脈內皮細胞(HUVECs)血管細胞間黏附分子-1(VCAM-1)表達的影響。方法原代培養(yǎng)HUVECs,2~3代進行試驗,MTT實驗設立空白對照組、Hcy(0.2~1.2 mmol/L)組、Rosu(0.5、1.0、2.0和5.0 mmol/L)組,實驗組:Rosu 1.0 mmol/L和2.0 mmol/L進行預處理細胞,2 h后加入0.6 mmol/L濃度的Hcy損傷HUVECs,Real time PCR法測定VCAM-1 mRNA表達,Western blot法測定細胞VCAM-1蛋白含量;Rosu 2.0 mmol/L+甲羥戊酸(Mev)100μmol/L預處理2 h后再加入0.6mmol/L濃度的Hcy以了解Mev對VCAM-1蛋白表達的影響。結果 Hcy組VCAM-1蛋白和mRNA表達較空白對照組明顯增強(P0.05);Rosu組VCAM-1蛋白和mRNA表達較Hcy組明顯減弱(P0.05),且Mev不能阻斷Rosu的這種作用。結論 Rosu可通過抑制HUVECs表達VCAM-1產生抗炎作用,且這種作用獨立于其調脂作用。
[Abstract]:Objective to investigate the effect of rosuvastatin on the expression of intercellular adhesion molecule-1 (VCAM-1) in human umbilical vein endothelial cells (HUVECs) injured by homocysteine (Hcy).Methods the primary culture of HUVECs2 ~ 2 ~ (2 +) was carried out by MTT assay. The control group was established as the control group, the control group was the control group, the control group was the control group, the control group was the control group, the control group was the control group, the control group was the control group, the control group was the control group, the control group was the control group.The cells of the experimental group were pretreated with 1. 0 mmol/L and 2. 0 mmol/L for 2 h, then the cells were pretreated with 0. 6 mmol/L concentration of Hcy for 2 h. The expression of VCAM-1 mRNA was measured by real time PCR method. Western blot method was used to determine the VCAM-1 protein content of the cells. Rosu 2. 0 mmol/L Mevedol 100 渭 mol/L was pretreated for 2 h, then 0.6mmol/L was added.Degree of Hcy to understand the effect of Mev on the expression of VCAM-1 protein.Results the expression of VCAM-1 protein and mRNA in Hcy group was significantly higher than that in control group. The expression of VCAM-1 protein and mRNA in Hcy group was significantly lower than that in Hcy group, and Mev could not block this effect of Rosu.Conclusion Rosu can produce anti-inflammatory effect by inhibiting HUVECs expression of VCAM-1, and this effect is independent of its lipid regulation.
