【摘要】：目的:探討利奈唑胺在腦膜炎患者腦脊液中的藥動學(xué)及藥效學(xué)參數(shù)變化。方法:10例重癥神經(jīng)外科患者入組,應(yīng)用利奈唑胺(600 mg/次,靜脈滴注,每日2次,每次靜脈滴注60 min)治療腦室外引流引起的革蘭陽性球菌感染所致的腦膜炎,當(dāng)藥物達(dá)穩(wěn)態(tài)后測定其在腦脊液中的藥動學(xué)參數(shù),結(jié)合微生物學(xué)評價其藥效學(xué)指標(biāo)。結(jié)果:利奈唑胺在用藥2.5 h達(dá)到峰濃度(cmax)(5.5±2.5)μg/ml,其谷濃度(cmin)是在用藥0和12 h時出現(xiàn),分別為(3.1±1.3)μg/ml和(3.3±1.8)μg/ml;AUC0-12 h為(105±59)μg·h/ml,性別對AUC0-12 h的影響差異無統(tǒng)計學(xué)意義[男性:(112±58)μg·h/ml,女性:(104±49)μg·h/ml,(P0.05)];藥物對腦脊液的平均穿透率為67%;最低抑菌濃度(MIC)≤2μg/ml時,AUC0-24 h/MIC的值為112.6,濃度2μg/ml的時間是(11.2±2.8)h,抗菌藥物超MIC時間(TMIC)占100%,90%MIC(MIC90)≤4μg/ml時,AUC0-24 h/MIC90的值為57.1,TMIC占54%。結(jié)論:利奈唑胺對腦膜炎患者腦脊液有良好的穿透率,常規(guī)劑量能達(dá)到有效治療濃度。
[Abstract]:Objective: to investigate the pharmacokinetic and pharmacodynamic parameters of linazolamide in cerebrospinal fluid (CSF) of patients with meningitis.Methods 10 patients with severe neurosurgery were treated with linazolamine 600 mg/, intravenous drip twice a day, each time for 60 minutes, in order to treat meningitis caused by Gram-positive cocci infection caused by ventricular drainage.The pharmacokinetic parameters of the drug in cerebrospinal fluid (CSF) were determined when the drug reached steady state, and its pharmacodynamics was evaluated with microbiology.Results: the peak concentration of linazolamide was 5.5 鹵2.5 渭 g / ml at 2.5 h, and its valley concentration was found at 0 h and 12 h after administration.鍒嗗埆涓，
