【摘要】：目的探討氯吡格雷對大鼠血小板聚集及肝細(xì)胞色素P4503A4表達(dá)的影響。方法 45只Wistar大鼠按照體重隨機(jī)分為3組(空白組,對照組和實驗組),每組15只�？瞻捉M大鼠灌胃生理鹽水5 m L·kg-1·d-1,對照組灌胃阿司匹林100 mg·kg-1·d-1,實驗組大鼠灌胃氯吡格雷20 mg·kg-1·d-1,均5m L,3組大鼠均處理7 d。藥物處理7 d后,心臟取血,檢測血小板聚集率和血小板聚集抑制率。Western blot法檢測3組肝細(xì)胞色素P4503A4表達(dá)水平。結(jié)果實驗組大鼠血小板最大聚集率為(37.23±5.01)%,顯著低于空白組的(57.21±5.23)%和對照組的(47.23±4.21)%(P0.05);實驗組大鼠血小板聚集抑制率為(33.56±4.64)%,顯著高于對照組的(21.12±3.36)%(P0.05);與空白組和對照組相比,實驗組大鼠肝細(xì)胞色素P4503A4表達(dá)水平顯著降低(P0.05)。結(jié)論氯吡格雷可能通過抑制大鼠肝細(xì)胞色素P4503A4表達(dá)來達(dá)到抗血小板功能的作用。
[Abstract]:Objective to investigate the effects of clopidogrel on platelet aggregation and hepatic cytochrome P4503A4 expression in rats.Methods 45 Wistar rats were randomly divided into 3 groups according to their body weight: blank group, control group and experimental group with 15 rats in each group.Rats in the blank group were given normal saline 5 mL kg-1 d-1, the control group was given aspirin 100mg kg-1 d-1, the experimental group was treated with clopidogrel 20 mg kg-1 d-1, and the rats in the control group were treated with clopidogrel 20 mg kg-1 d-1 for 7 days.After 7 days of treatment, blood was taken from the heart, platelet aggregation rate and platelet aggregation inhibition rate.
