液相色譜-串聯(lián)質(zhì)譜法測定人血漿中奧美拉唑的血藥濃度及其藥動學(xué)

【摘要】:目的:建立液相色譜-串聯(lián)質(zhì)譜法測定人血漿中奧美拉唑的血藥濃度并進行藥動學(xué)研究。方法:以蘭索拉唑為內(nèi)標,血漿樣品經(jīng)乙腈沉淀后,經(jīng)LC-MS/MS分離分析。采用Diamonsil C18柱(2.1 mm×150 mm,5μm),甲醇-水(含0.01%甲酸)(67∶33)為流動相;流速為0.3 ml·min-1,采用電噴霧離子源(ESI),以多反應(yīng)監(jiān)測方式(MRM)進行負離子監(jiān)測,奧美拉唑和蘭索拉唑的定量分析離子對分別為m/z 344.2/193.8,367.9/163.8。結(jié)果:奧美拉唑在3.38~2 110.00ng·ml-1(r=0.997 1)范圍內(nèi)線性關(guān)系良好,最低定量限為3.38 ng·ml-1,方法回收率在92.48%~99.39%,日內(nèi)(n=5)RE均小于3.96%,日間(n=15)RE均小于6.41%。結(jié)論:該方法快速簡便,靈敏準確,可用于奧美拉唑血藥濃度監(jiān)測和藥動學(xué)研究。
[Abstract]:Objective: to establish a liquid chromatography-tandem mass spectrometry (LC-MS) method for the determination of omeprazole in human plasma and its pharmacokinetics.Methods: Lansoprazole was used as internal standard.
