EGF類生長(zhǎng)因子來(lái)源的新型靶向肽在抗腫瘤藥物蛋白中的應(yīng)用

【摘要】:許多腫瘤細(xì)胞表面表皮生長(zhǎng)因子受體EGFR都存在過(guò)表達(dá)現(xiàn)象?疾炝伺6徊《旧L(zhǎng)因子(VGF)中的EGFR結(jié)合域(S3)與人的肝素樣表皮生長(zhǎng)因子(HB-EGF)來(lái)源的肝素結(jié)合域(命名為HE)重組后對(duì)腫瘤細(xì)胞的選擇性。通過(guò)重組表達(dá)帶有靶向和穿膜結(jié)構(gòu)域的EGFP-S3-HE和EGFP-S3-HE-TATm兩種融合蛋白與正常細(xì)胞和腫瘤細(xì)胞的共孵育實(shí)驗(yàn)來(lái)研究其對(duì)腫瘤細(xì)胞的特異性靶向吸附和穿膜效應(yīng)。進(jìn)一步將S3-HE-TATm靶向穿膜序列與苦瓜來(lái)源的核糖體失活蛋白MAP30融合,可顯著提高M(jìn)AP30對(duì)腫瘤細(xì)胞的抑殺作用,但這種抑殺作用卻對(duì)正常細(xì)胞仍保持在較低水平。由此表明S3-HE-TATm是一種新型優(yōu)異的腫瘤細(xì)胞靶向藥物運(yùn)輸載體,可用于腫瘤治療的進(jìn)一步開(kāi)發(fā)研究。
[Abstract]:The expression of epidermal growth factor receptor (EGFR) on the surface of many tumor cells was investigated. The EGFR binding domain S3 of cowpox virus growth factor (VGF) and heparin binding domain from human heparin-like epidermal growth factor (HB-EGF) were investigated. The specific target for tumor cells was studied by co-incubating EGFP-S3-HE and EGFP-S3-HE-TATm fusion proteins with targeted and transmembrane domains with normal cells and tumor cells. Further fusion of S3-HE-TATm targeted transmembrane sequence with ribosomal inactivated protein MAP30 derived from Momordica charantia L. The inhibitory effect of MAP30 on tumor cells was significantly increased, but the inhibitory effect on normal cells remained at a low level, which indicated that S3-HE-TATm was a novel and excellent drug carrier for tumor cell targeting. It can be used in the further development of tumor therapy.
