1株抗腫瘤銀杏內(nèi)生真菌的發(fā)酵條件及活性產(chǎn)物穩(wěn)定性研究

【摘要】:對前期研究發(fā)現(xiàn)的1株分離自銀杏樹皮組織的具有良好的抗腫瘤活性內(nèi)生真菌米曲霉菌(Aspergillus oryzae)YX-5菌株的發(fā)酵條件及活性產(chǎn)物穩(wěn)定性進行研究。以發(fā)酵后的菌體干質(zhì)量、粗提物質(zhì)量和粗提物抗腫瘤活性為指標(biāo),對YX-5菌株的發(fā)酵溫度、pH值、轉(zhuǎn)速和發(fā)酵時間等條件進行優(yōu)化。結(jié)果表明,最適宜的發(fā)酵條件為溫度25℃、pH值7.0、搖床轉(zhuǎn)速110 r/min、發(fā)酵時間7 d。以粗提物的抗腫瘤活性為指標(biāo),比較發(fā)酵液粗提物經(jīng)熱處理、酸堿處理、紫外照射和蛋白酶處理后的穩(wěn)定性,發(fā)現(xiàn)YX-5菌株的活性產(chǎn)物在常溫、中性和酸性環(huán)境中穩(wěn)定性較好,對紫外線和蛋白酶有較強的抗性,但在超過100℃高溫條件下或pH值超過9.0的堿性環(huán)境中穩(wěn)定性較差。該菌株粗提物在自然條件下總體穩(wěn)定性較高,不易發(fā)生變化,有利于活性成分分離工作的開展。
[Abstract]:The fermentation conditions and the stability of the active product of an endophytic fungus Aspergillus oryzae oryzae)YX-5, which was isolated from the bark of Ginkgo, were studied.
