曲伏前列素致高眼壓模型大鼠結(jié)膜充血的不良反應(yīng)機(jī)制研究

【摘要】:目的:研究曲伏前列素致高眼壓模型大鼠結(jié)膜充血的不良反應(yīng)機(jī)制。方法:將50只大鼠隨機(jī)分為正常對照組、模型組和曲伏前列素低、中、高劑量組(100、200、400 mg/kg),每組10只。除正常對照組外,其余各組大鼠右眼建立高眼壓模型,每天滴加相應(yīng)藥物1次,連續(xù)1周。末次給藥后體外分離并培養(yǎng)各組大鼠右眼角膜周圍血管內(nèi)皮細(xì)胞,檢測血管內(nèi)皮細(xì)胞活力、細(xì)胞凋亡情況和增殖相關(guān)因子(Ki-76)、凋亡相關(guān)因子(Bad、Bax)、凋亡抑制相關(guān)因子(Bcl-2、Bcl-xl)蛋白表達(dá)水平。結(jié)果:與正常對照組比較,模型組大鼠血管內(nèi)皮細(xì)胞活力明顯降低,凋亡率明顯增加,Bad、Bax蛋白表達(dá)明顯增強(qiáng),Bcl-2、Bcl-xl、Ki-76蛋白表達(dá)明顯減弱,以上差異均有統(tǒng)計(jì)學(xué)意義(P0.05或P0.01)。與模型組比較,曲伏前列素低、中、高劑量組大鼠血管內(nèi)皮細(xì)胞活力明顯降低,凋亡率明顯增加,Bcl-2、Bcl-xl蛋白表達(dá)明顯減弱,以上差異均有統(tǒng)計(jì)學(xué)意義(P0.05或P0.01);曲伏前列素中、高劑量組大鼠血管內(nèi)皮細(xì)胞中Bad、Bax蛋白表達(dá)明顯增強(qiáng)(P0.05或P0.01),其余指標(biāo)差異均無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:曲伏前列素可能是通過促進(jìn)高眼壓模型大鼠角膜周圍血管內(nèi)皮細(xì)胞凋亡,從而降低其細(xì)胞活力,進(jìn)而引發(fā)結(jié)膜充血這一不良反應(yīng)。
[Abstract]:Objective: to study the adverse reaction mechanism of conjunctival hyperemia induced by travoprostatin in rats. Methods: 50 rats were randomly divided into normal control group, model group and travoprostatin low, middle and low. 10 rats in each group, except the normal control group, established a model of intraocular hypertension in the right eye of the other groups, and the rats were given corresponding drugs once a day. After the last administration of the drug in vitro, vascular endothelial cells were isolated and cultured around the cornea of the right eye of the rats in each group, and the activity of vascular endothelial cells was detected. The expression level of apoptosis and proliferation-related factors Ki-76, Bad Baxan, Bcl-2 Bcl-xl) protein. Results: compared with the normal control group, the activity of vascular endothelial cells in the model group was significantly lower than that in the normal control group, and the expression of Bcl-2Bcl-xl protein in the model group was significantly lower than that in the control group. Apoptosis rate significantly increased the expression of Bad-Bax protein increased significantly, and the expression of Bcl-2Bcl-xlP1-Ki-76 protein decreased significantly. Compared with the model group, the expression of travoprostatin was lower than that of the model group, and the activity of vascular endothelial cells in the medium and high dose groups was significantly lower than that in the model group. The expression of Bcl-xl protein decreased significantly, and the above differences were statistically significant (P0.05 or P0.01A), and the expression of Bcl-xl protein in travoprostatin was significantly lower than that in control group (P 0.05 or P 0.01). In high dose group, the expression of Bad-Bax protein in vascular endothelial cells increased significantly (P 0.05 or P 0.01), but there was no significant difference in other indexes (P 0.05). Conclusion: trafoprostatin may promote apoptosis of endothelial cells around cornea of rats with high intraocular pressure. Thus reduce its cell viability, and then cause the adverse reaction of conjunctival congestion.
