【摘要】：目的觀察小檗堿對糖尿病腎病大鼠腎臟足細(xì)胞相關(guān)蛋白nephrin、podocin和α3β1整合素蛋白表達(dá)的影響,探討小檗堿對糖尿病腎病大鼠腎臟的保護(hù)作用及部分機(jī)制。方法采用高糖高脂飼料喂養(yǎng)6周,聯(lián)合低劑量鏈脲佐菌素誘導(dǎo)糖尿病腎病大鼠模型。隨機(jī)分為正常對照組、模型組、3個不同劑量(50、100和200 mg·kg-1)小檗堿給藥組和依那普利陽性藥對照組(1 mg·kg-1)。分別采用免疫組織化學(xué)法,油鏡(×1 000倍)和高倍鏡(×400倍)和Western blot法,觀察和檢測腎臟足細(xì)胞相關(guān)蛋白nephrin、podocin及α3β1整合素的分布和表達(dá)水平。結(jié)果足細(xì)胞相關(guān)蛋白nephrin、podocin及α3β1整合素蛋白主要分布于足細(xì)胞,但分布位置略有差異;與正常對照組相比,模型組大鼠的足細(xì)胞相關(guān)蛋白nephrin、podocin及α3β1整合素的表達(dá)水平明顯降低;與模型組相比,小檗堿(100和200 mg·kg-1)給藥組明顯改善糖尿病腎病大鼠腎臟形態(tài)學(xué)異常,上調(diào)足細(xì)胞相關(guān)蛋白nephrin、podocin及α3β1整合素的表達(dá)水平。結(jié)論小檗堿可以緩解糖尿病腎病大鼠腎臟病理異常改變及蛋白尿的產(chǎn)生,這可能與其上調(diào)足細(xì)胞相關(guān)蛋白nephrin、podocin及整合素α3β1的表達(dá)相關(guān)。
[Abstract]:Objective to observe the effect of berberine on the expression of nephrinpodocin and 偽 3 尾 1 integrin protein in renal podocin of diabetic nephropathy rats. To investigate the protective effect of berberine on the kidney of diabetic nephropathy rats and its mechanism. Methods the diabetic nephropathy rats were fed with high glucose and high fat diet for 6 weeks and induced by low dose streptozotocin. The rats were randomly divided into normal control group. Model group, three different doses of berberine 50 100 and 200 mg kg-1) and enalapril positive control group (1 mg kg -1) were treated with immunohistochemical method, oil microscope (脳 1 000 times), high power microscope (脳 400 times) and Western blot method, respectively. The distribution and expression level of nephrinpodocin and 偽 3 尾 1 integrin in kidney podocin were observed and detected. Results the nephrinpodocin and 偽 3 尾 1 integrin proteins were mainly distributed in podocins, but there were some differences in the distribution of nephrinpodocin and 偽 3 尾 1 integrin. The expression of podocin and 偽 3 尾 1 integrin in podocin and 偽 3 尾 1 in the model group was significantly decreased, compared with the model group, berberine 100 and 200 mg kg-1) significantly improved the renal morphology abnormality in diabetic nephropathy rats. The expression of nephrininpodocin and 偽 3 尾 1 integrin was up-regulated. Conclusion berberine can attenuate the pathological changes of kidney and the production of proteinuria in diabetic nephropathy rats. This may be related to its up-regulation of the expression of podocin and integrin 偽 3 尾 1.
