尿酸酶-過氧化氫酶脂質體的特性分析及藥效學研究

【摘要】:目的考察尿酸酶-過氧化氫酶脂質體(uricase and catalase liposome,UCALP)、游離尿酸酶(uricase,UAE)的最適溫度和最適pH,并探究UCALP降低高尿酸血癥小鼠的尿酸水平和過氧化氫水平的能力。方法采用逆向蒸發(fā)法制備UCALP,并分別測定UCALP和UAE的最適溫度和最適pH;小鼠腹腔注射尿酸建立高尿酸血癥模型后,分別靜脈注射UCALP和UAE,用測試盒分別測定不同時間點小鼠血清中尿酸濃度和過氧化氫的濃度。結果 UCALP和UAE的最適溫度均為40℃,最適pH分別為8.0和8.5;在高尿酸血癥的小鼠體內,UCALP比UAE降尿酸水平更明顯,且UCALP組小鼠體內的過氧化氫濃度較UAE組低。結論UCALP能有效降低高尿酸血癥小鼠的尿酸水平,且能在一定程度上降低過氧化氫的濃度。
[Abstract]:Objective to investigate the optimal temperature and pH of uric acidase-catalase liposome and catalase liposome UCALPe, and to explore the ability of UCALP to reduce the level of uric acid and hydrogen peroxide in hyperuricemia mice. The optimal temperature and pH of UCALP and UAE were determined by evaporation, and the hyperuricemia model was established by intraperitoneal injection of uric acid in mice. UCALP and UAE were injected intravenously. The serum uric acid concentration and hydrogen peroxide concentration in mice were measured with test box at different time points.
