牛膝有效部位在腫瘤微環(huán)境下促進(jìn)樹(shù)突狀細(xì)胞成熟及功能調(diào)控的研究

【摘要】:目的觀察牛膝有效部位在腫瘤微環(huán)境下對(duì)樹(shù)突狀細(xì)胞成熟的促進(jìn)及功能調(diào)控作用。方法干燥牛膝粉末提取并鑒定牛膝總皂苷和牛膝多糖;建立模擬腫瘤微環(huán)境的MCF-7乳腺癌細(xì)胞與樹(shù)突狀細(xì)胞共培養(yǎng)體系,并分別給予牛膝多糖、牛膝總皂苷及牛膝多糖和總皂苷協(xié)同刺激;流式細(xì)胞儀檢測(cè)樹(shù)突狀細(xì)胞表面分子的表達(dá)及樹(shù)突狀細(xì)胞誘導(dǎo)的細(xì)胞毒性T淋巴細(xì)胞(CTL)殺傷活性,ELISA法檢測(cè)培養(yǎng)上清中白細(xì)胞介素-1β、腫瘤壞死因子-α、白細(xì)胞介素-10和白細(xì)胞介素-12的水平,Western blot檢測(cè)JAK/STAT3信號(hào)通路中p-JAK、JAK、p-STAT3、STAT3的蛋白表達(dá)。結(jié)果在腫瘤微環(huán)境下,牛膝有效部位牛膝總皂苷和牛膝多糖均可以通過(guò)TLR4及Dectin-1受體途徑,啟動(dòng)樹(shù)突狀細(xì)胞細(xì)胞JAK/STAT3信號(hào)通路,上調(diào)樹(shù)突狀細(xì)胞細(xì)胞表面分子CD80、CD86及CD40的表達(dá),從而誘導(dǎo)樹(shù)突狀細(xì)胞的成熟與分化,促進(jìn)腫瘤壞死因子-α、白細(xì)胞介素-12、白細(xì)胞介素-10及白細(xì)胞介素-1β等細(xì)胞因子的分泌,兩者協(xié)同較單獨(dú)應(yīng)用作用加強(qiáng)。結(jié)論以適當(dāng)形式的中藥有效部位作為免疫增強(qiáng)劑聯(lián)合樹(shù)突狀細(xì)胞疫苗,可有效糾正腫瘤微環(huán)境下的免疫缺陷,有效激活腫瘤特異性CTL細(xì)胞,從而產(chǎn)生特異性抗腫瘤效應(yīng)。
[Abstract]:Objective to observe the effects of active fractions of Achyranthes bidentata on the maturation and function of dendritic cells in tumor microenvironment. Methods the total saponins and polysaccharides of Achyranthes bidentata were extracted and identified by dried powder of Achyranthes bidentata. To establish a co-culture system of MCF-7 breast cancer cells and dendritic cells simulated by tumor microenvironment, and give them co-stimulation of Achyranthes bidentata polysaccharides, total saponins, Achyranthes bidentata polysaccharides and total saponins, respectively. Flow cytometry was used to detect the expression of dendritic cell surface molecules and the cytotoxic T lymphocyte CTL cytotoxicity induced by dendritic cells. Elisa was used to detect interleukin-1 尾, tumor necrosis factor- 偽 and interleukin-10 in the culture supernatant. The levels of IL-12 and IL-12 were measured by Western blot to detect the protein expression of p-JAKT3-STAT3STAT3 in the JAK/STAT3 signaling pathway. The total saponins of Achyranthes bidentata and polysaccharides of Achyranthes bidentata can activate the JAK/STAT3 signaling pathway of dendritic cells through TLR4 and Dectin-1 receptor pathway and up-regulate the expression of CD80 CD86 and CD40 on the surface of dendritic cells. Thus inducing the maturation and differentiation of dendritic cells and promoting the secretion of cytokines such as tumor necrosis factor- 偽, interleukin-12, interleukin-10 and interleukin-1 尾. Conclusion the effective part of traditional Chinese medicine combined with dendritic cell vaccine can effectively correct the immune deficiency in tumor microenvironment and activate tumor specific CTL cells. Thus, the specific anti-tumor effect is produced.
