索拉菲尼對骨肉瘤細胞的抑制作用及分子機制研究

【摘要】:目的研究索拉菲尼對骨肉瘤細胞增殖和凋亡的作用及其分子機制。方法將人骨肉瘤細胞(HOS細胞)分為對照組及索拉菲尼組,用3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴鹽比色法觀察索拉菲尼對HOS細胞活力的影響,用蛋白質印跡法檢測凋亡相關蛋白變化,用Real-time PCR檢測let-7d變化情況。結果索拉菲尼可抑制HOS細胞活力,并呈劑量和濃度依賴性;同時,索拉菲尼可以抑制Lin28表達,同時促進let-7d表達;過表達Lin28可以逆轉索拉菲尼的促凋亡作用。結論索拉菲尼通過調控Lin28/le-7d環(huán)路來促進骨肉瘤細胞凋亡。
[Abstract]:Objective to study the effect of Solafenil on proliferation and apoptosis of osteosarcoma cells and its molecular mechanism. Methods Human osteosarcoma cell line HOS cells were divided into control group and Solafenil group. The effects of Solafenil on the activity of HOS cells were observed by using 3-tiaphane 5-dimethylthiazole-2-triazolium 5-diphenyl tetrazolium bromide colorimetry, and the changes of apoptosis-related proteins were detected by Western blotting. Real-time PCR was used to detect the changes of let-7d. Results Solafinil inhibited the activity of HOS cells in a dose-and dose-dependent manner, and at the same time, it inhibited the expression of Lin28 and promoted the expression of let-7d. Overexpression of Lin28 can reverse the apoptosis-promoting effect of Solafini. Conclusion Solafini can promote the apoptosis of osteosarcoma cells by regulating the Lin28/le-7d loop.
