【摘要】：目的觀察五步蛇毒蛋白C激活物(PCA)對人臍靜脈血管內(nèi)皮細胞(HUVEC)的保護作用,并分析其對組織因子(TF)表達的影響。方法常規(guī)培養(yǎng)HUVEC,實驗分為空白對照組、脂多糖(LPS)組、PCA(0.625、1.25、2.50μg·mL~(-1))組和PCA+LPS組。MTT法檢測HUVEC活性,ELISA檢測HUVEC培養(yǎng)上清中TF分泌量,免疫熒光染色檢測核轉(zhuǎn)錄因子(NF)-κB是否被激活-核轉(zhuǎn)運。結(jié)果 PCA對HUVEC活性和形態(tài)無顯著影響(P0.05)。與空白對照組,LPS組細胞活性顯著下降(P0.01);而LPS+PCA組細胞活性顯著高于LPS組(P0.05)。LPS組TF表達量和NF-κB表達均顯著高于空白對照組(P0.01);LPS+PCA組與LPS組比較TF表達量和NF-κB表達降低(P0.01)。結(jié)論 PCA可降低HUVEC分泌TF,減少LPS對HUVEC的損傷作用,其可能與抑制NF-κB通路的活化有關(guān)。
[Abstract]:Objective to observe the protective effect of PCA on human umbilical vein endothelial cells (HUVECs) and to analyze the effect of PCAs on the expression of tissue factor TF.Methods HUVECs were routinely cultured and divided into blank control group. Lipopolysaccharide (LPS) group and PCA LPS group were used to detect the activity of HUVEC and Elisa to detect TF secretion in the supernatant of HUVEC culture. Results PCA had no significant effect on the activity and morphology of HUVEC. Compared with the control group, the activity of NF-kappa B was significantly decreased, while the activity of LPS PCA group was significantly higher than that of LPS PCA group (P < 0.05), while that of LPS PCA group was significantly higher than that of LPS PCA group (P < 0.01), while that of LPS PCA group was significantly higher than that of LPS PCA group (P < 0.05). The expression of TF and NF- 魏 B in the LPS group was significantly higher than that in the control group (P 0.01) and the expression of NF- 魏 B in the LPS group was lower than that in the LPS group. Conclusion PCA can decrease the secretion of HUVEC and decrease the damage of LPS to HUVEC. It may be related to inhibiting the activation of NF- 魏 B pathway.
