【摘要】：以香草酸為母體,分別與6-氨基己酸(EACA)、止血芳酸(AMBA)和止血環(huán)酸(AMCA)反應(yīng),合成3種未見文獻(xiàn)報道的香草酸酰胺衍生物,并采用IR、ESI-MS、1H NMR和13C NMR進(jìn)行結(jié)構(gòu)鑒定。以傳統(tǒng)方法、熒光動力學(xué)法和紫外動力學(xué)法測定其對血漿復(fù)鈣時間(PRT)的影響。為了研究衍生物在血液中的穩(wěn)定性,在模擬生理條件下,運用熒光光譜、紫外光譜及分子對接技術(shù)研究了它分別與人血清白蛋白(HSA)的結(jié)合。結(jié)果表明,3種衍生物在一定濃度范圍內(nèi)都可以有效縮短PRT,具有顯著的止血活性。新的動力學(xué)測定方法,不僅有效避免了傳統(tǒng)方法引入的人為誤差,重現(xiàn)性良好,而且判斷標(biāo)準(zhǔn)更加科學(xué),并可以獲得更多血漿凝結(jié)過程中的信息。3種衍生物均主要通過氫鍵和范德華力與HSA結(jié)合形成穩(wěn)定的復(fù)合物,表明它可通過HSA在血液中傳遞并運輸?shù)较鄳?yīng)部位而發(fā)生藥理作用。
[Abstract]:Three vanilamide derivatives were synthesized from vanillic acid by reacting with 6-amino-caproic acid (EACAA), hemostatic aromatic acid (AMBA) and hemostatic acid (AMCA) respectively. The structures of vanilamide derivatives were identified by IR ESI-MS 1H NMR and 13C NMR. In order to study the stability of derivatives in blood, fluorescence spectra were used under simulated physiological conditions. UV spectra and molecular docking techniques were used to study the binding of HSA with human serum albumin. The results showed that the three derivatives could effectively shorten PRT in a certain concentration range, and had remarkable hemostatic activity. It not only effectively avoids the artificial error introduced by the traditional method, but also has good reproducibility, and the judgment standard is more scientific. More information in plasma coagulation can be obtained. 3 derivatives can form stable complexes by hydrogen bonding and van der Waals force binding with HSA, which indicates that they can be transported to the corresponding sites through HSA and have pharmacological effects.
