【摘要】:為進(jìn)一步提高林可霉素生產(chǎn)發(fā)酵水平,本文以林肯鏈霉菌LK15-35為出發(fā)菌株,采用氯化鋰(Li Cl)、常壓室溫等離子體(ARTP)和紫外線(UV)三重復(fù)合誘變,首先用Li Cl預(yù)處理4h,然后用ARTP照射60s,再用UV照射30s,最終獲得高產(chǎn)突變株LK16-77。結(jié)果表明,搖瓶發(fā)酵單位提高30.0%,經(jīng)180噸罐生產(chǎn)驗(yàn)證,平均放罐效價(jià)提高21.4%。穩(wěn)定性傳代驗(yàn)證證明該突變株高產(chǎn)性能遺傳穩(wěn)定。
[Abstract]:In order to further improve the fermentation level of lincomycin, Streptomyces Lincoln LK15-35 was used as the starting strain. The high yield mutant LK16-77 was obtained by pretreatment with Li Cl for 4 h, then irradiated with ARTP for 60 s and then irradiated with UV for 30 s. The results showed that the fermentation unit of shaking flask was increased by 30.0% and verified by 180 ton pot production. The average titer of the mutant was increased by 21.4.The stability test proved that the mutant was genetically stable in high yield.
