鮭魚降鈣素磷脂復(fù)合物的制備及質(zhì)量評價

【摘要】:目的:研究鮭魚降鈣素磷脂復(fù)合物(s CT-PC)的制備工藝并對其進行質(zhì)量評價。方法:以s CTPC的復(fù)合率為評價指標(biāo),并用單因素試驗和正交設(shè)計試驗[L16(4)5]進行處方優(yōu)化;研究s CT-PC的油水分配系數(shù)、紅外特征、晶體特征及相變特性等理化特性。結(jié)果:s CT-PC的最佳制備條件為鮭魚降鈣素(s CT)與磷脂(PC)的質(zhì)量比為1∶15,反應(yīng)溶劑為二氯甲烷-乙醚(V/V=6∶4),反應(yīng)時間與溫度分別為25℃和4 h;s CT-PC在正辛醇中的溶解度及油水分配系數(shù)較s CT或s CT與PC的物理混合物均明顯提高;紅外分光光度法(IR)、X-射線衍射法(XRD)、差示掃描熱量法(DSC)均驗證該復(fù)合物的形成。結(jié)論:s CT-PC的形成受反應(yīng)溶劑、反應(yīng)時間及藥脂比影響較大;s CT-PC的親脂性較s CT明顯增強,為進一步制備鮭魚降鈣素磷脂復(fù)合物的納米粒奠定基礎(chǔ)。
[Abstract]:Objective: to study the preparation process of salmon calcitonin phospholipid complex (sCT-PC) and evaluate its quality. Methods: the compound ratio of s CTPC was used as evaluation index, and the formulation was optimized by single factor test and orthogonal design test. The oil / water partition coefficient and infrared characteristics of s CT-PC were studied.
