沒食子酸乙酯通過線粒體通路誘導(dǎo)人結(jié)腸癌Lovo細(xì)胞凋亡的機(jī)制研究

【摘要】:目的研究沒食子酸乙酯(ethyl gallate,EG)對(duì)人結(jié)腸癌細(xì)胞Lovo凋亡的影響并探討相關(guān)機(jī)制。方法以體外培養(yǎng)的Lovo細(xì)胞作為研究對(duì)象,采用倒置顯微鏡觀察細(xì)胞生長的變化、Annexin V-FITC/PI雙染法和DNA Ladder瓊脂糖凝膠電泳法檢測(cè)沒食子酸乙酯的凋亡誘導(dǎo)作用;采用免疫細(xì)胞化學(xué)法檢測(cè)cleaved caspase-3,cleaved caspase-9蛋白的表達(dá)。結(jié)果沒食子酸乙酯可明顯抑制Lovo細(xì)胞生長,倒置顯微鏡下觀察到細(xì)胞密度明顯降低:各用藥組的細(xì)胞凋亡率隨著藥物濃度升高而升高:DNA瓊脂糖凝膠電泳可見明顯DNA梯狀條帶:各用藥組細(xì)胞cleaved caspase-3,cleaved caspase-9基因蛋白陽性表達(dá)率明顯升高。結(jié)論沒食子酸乙酯可通過上調(diào)cleaved caspase-3和cleaved caspase-9蛋白的表達(dá),激活線粒體通路從而促進(jìn)Lovo細(xì)胞凋亡,這是沒食子酸乙酯發(fā)揮其抗癌作用的重要機(jī)制之一。
[Abstract]:Objective to study the effect of ethyl gallate EGG on apoptosis of human colon cancer cell line Lovo and its mechanism. Methods Lovo cells were cultured in vitro. The changes of cell growth were observed by inverted microscope. Annexin V-FITC / Pi double staining and DNA Ladder agarose gel electrophoresis were used to detect the apoptosis-inducing effect of ethyl gallate. The expression of cleaved caspase-3 cleaved caspase-9 protein was detected by immunocytochemistry. Results Ethyl gallate could significantly inhibit the growth of Lovo cells. Under the inverted microscope, the cell density decreased obviously: the apoptosis rate of each drug group increased with the increase of drug concentration, and the DNA ladder was observed by agarose gel electrophoresis. The cell cleaved caspase-3 cleaved caspase-9 gene protein in each medication group was also observed. Conclusion Ethyl gallate can up-regulate the expression of cleaved caspase-3 and cleaved caspase-9 protein. Activation of mitochondrial pathway to promote apoptosis of Lovo cells is one of the important mechanisms by which ethyl gallate exerts its anticancer effect.
