羅格列酮對慢性阻塞性肺疾病大鼠的抗炎作用

【摘要】:目的研究羅格列酮對慢性阻塞性肺疾病(COPD)大鼠肺組織勻漿中白細胞介素-6(IL-6)、基質(zhì)金屬蛋白酶-2(MMP-2)、基質(zhì)金屬蛋白酶-9(MMP-9)、腫瘤壞死因子-α(TNF-α)的影響。方法采用氣管內(nèi)滴注脂多糖聯(lián)合香煙煙熏建立COPD大鼠模型。SPF級成年雄性Wistar大鼠50只隨機分為5組,對照組(A組)、模型組(B組)、羅格列酮組1(C組)、羅格列酮組2(D組)、羅格列酮組3(E組),每組10只,C組、D組、E組每天上午煙熏前分別給予羅格列酮鈉片0.1、0.15、0.2 mg/kg灌胃,A組、B組給予1 m L生理鹽水灌胃,30 d后觀察大鼠的一般狀況和體重改變。采用酶聯(lián)免疫吸附法(ELISA)檢測肺組織勻漿中IL-6、MMP-2、MMP-9及TNF-α含量,觀察羅格列酮對COPD大鼠的抗炎作用。結(jié)果實驗開始前所測的各組大鼠體重差異無統(tǒng)計學意義(P0.05)。實驗結(jié)束時,與A組體重變化相比,B組大鼠體重增長最少,C組、D組、E組次之(P0.05)。C、D、E組三組大鼠之間體重變化差異無統(tǒng)計學意義(P0.05)。B組肺組織勻漿中MMP-2、TNF-α的表達均較A、C、D、E組明顯升高(P0.05),C、D、E組中MMP-2、TNF-α的表達較A組升高(P0.05),較B組減低(P0.01),3組間差異無統(tǒng)計學意義(P0.05)。B組肺組織勻漿中MMP-9、IL-6的含量最高,A組最低,C、D、E組隨著羅格列酮濃度的升高MMP-9、IL-6的含量逐漸降低,差異有統(tǒng)計學意義(P0.05)。結(jié)論羅格列酮對抑制肺部炎癥介質(zhì)的釋放、基質(zhì)金屬蛋白酶的產(chǎn)生具有重要作用,可減輕氣道炎癥、肺實質(zhì)損傷和氣道重塑。羅格列酮在COPD的防治中具有一定的作用。
[Abstract]:Objective to study the effects of rosiglitazone on interleukin-6 (IL-6), matrix metalloproteinase-2 (MMP-2), matrix metalloproteinase-9 (MMP-9), tumor necrosis factor- 偽 (TNF- 偽) in lung tissue homogenate of rats with chronic obstructive pulmonary disease (COPD). Establishment of COPD rat model by combination of sugar and cigarette smoke. Fifty male Wistar rats of SPF grade were randomly divided into 5 groups. Control group, model group, rosiglitazone group, rosiglitazone group, rosiglitazone group, rosiglitazone group, rosiglitazone group, rosiglitazone group and rosiglitazone group, 10 rats in each group were given rosiglitazone sodium tablets 0.10.150.2 mg/kg before smoking every morning. The general status and weight change of rats were observed after oral administration of 1 mL normal saline for 30 days. The levels of IL-6, MMP-2, MMP-9 and TNF- 偽 in lung tissue homogenate were detected by enzyme linked immunosorbent assay (Elisa). To observe the anti-inflammatory effect of rosiglitazone on COPD rats. Results there was no significant difference in body weight of each group before the beginning of the experiment. Compared with the weight change of group A, the weight gain of rats in group B was the least than that in group C, group D, group E, followed by group E, there was no significant difference in body weight change among the three groups. There was no significant difference in the expression of MMP-2 and TNF- 偽 in lung tissue homogenate of group A and B, which were significantly higher than those of group ACDE in the group of P0.05TNF- 偽, and the expression of MMP-2TNF- 偽 in the lung tissue homogenate of group B was significantly higher than that in group ACDE. The expression of MMP-2 / TNF- 偽 was higher than that of group A (P 0.05A), but there was no significant difference between group B and group B (P 0.05). The content of MMP-9 and IL-6 in lung homogenate of group A and group A decreased gradually with the increase of rosiglitazone concentration, and the level of MMP-9IL-6 in group A decreased gradually with the increase of rosiglitazone concentration. Conclusion rosiglitazone plays an important role in inhibiting the release of pulmonary inflammatory mediators and producing matrix metalloproteinases, and can reduce airway inflammation. Lung parenchyma injury and airway remodeling. Rosiglitazone plays a role in the prevention and treatment of COPD.
