【摘要】：目的:以C48/80為陽(yáng)性藥物,比較3種細(xì)胞系(RBL-2H3,P8l5和Ku812細(xì)胞)及其指標(biāo)在評(píng)價(jià)藥物類過(guò)敏中的敏感性;并采用聚山梨酯80對(duì)3種細(xì)胞模型評(píng)價(jià)類過(guò)敏反應(yīng)的適用性進(jìn)行考察。方法:以不同濃度的C48/80分別作用于3種細(xì)胞系30和60 min,測(cè)定β-氨基糖苷酶釋放率、Annexin V陽(yáng)性細(xì)胞率以及IL-4和TNF-α釋放量。然后,用聚山梨酯80進(jìn)行模型的適用性考察,同時(shí)對(duì)不同濃度的聚山梨酯80引起的類過(guò)敏反應(yīng)進(jìn)行評(píng)價(jià)。結(jié)果:C48/80可引起3種細(xì)胞系的β-氨基糖苷酶釋放率、Annexin V陽(yáng)性細(xì)胞率、IL-4釋放量和TNF-α釋放量呈劑量依賴性增加;除了RBL-2H3細(xì)胞,其他2個(gè)細(xì)胞系檢測(cè)到的IL-4釋放量非常少,且3種細(xì)胞系檢測(cè)到的TNF-a量亦非常少;Annexin V陽(yáng)性細(xì)胞率的增加程度較其他3項(xiàng)指標(biāo)明顯;在相同刺激條件下,P815和Ku812細(xì)胞的β-氨基糖苷酶釋放率和Annexin V陽(yáng)性細(xì)胞率的突增較RBL-2H3細(xì)胞發(fā)生時(shí)間早,突增效果更明顯。聚山梨酯80的結(jié)果與C48/80給藥時(shí)基本吻合。此外,還發(fā)現(xiàn)聚山梨酯80≥0.05 mg·m L~(-1)時(shí),即存在引起類過(guò)敏反應(yīng)的風(fēng)險(xiǎn)。結(jié)論:β-氨基己糖苷酶釋放率和Annexin V陽(yáng)性細(xì)胞率是評(píng)價(jià)類過(guò)敏反應(yīng)的2種穩(wěn)定、可靠、重復(fù)性好的檢測(cè)指標(biāo);IL-4釋放量和TNF-α釋放量較適合作為輔助檢測(cè)指標(biāo);Annexin V陽(yáng)性率可較敏感地、較早地提示類過(guò)敏反應(yīng)的發(fā)生。P815和Ku812細(xì)胞較RBL-2H3細(xì)胞更適合作為一種早期、穩(wěn)定、敏感的肥大細(xì)胞脫顆粒體外檢測(cè)模型,用于致類過(guò)敏反應(yīng)藥物的早期篩選。
[Abstract]:Objective: to compare the sensitivity of three cell lines (RBL-2H3P8l5 and Ku812) with C48 / 80 in evaluating drug allergy. The applicability of polysorbate 80 to three cell models to evaluate the anaphylaxis reaction was investigated. Methods: three cell lines were treated with different concentrations of C48 / 80 for 30 min and 60 min, respectively. The release rate of 尾-aminoglycosidase and Annexin V positive rate were measured. Cell rate and release of IL-4 and TNF- 偽. Then, The applicability of the model was investigated with polysorbate 80. At the same time, the anaphylaxis reaction induced by different concentrations of polysorbate 80 was evaluated. Results the release rate of 尾 -aminoglycosidase and the release rate of Annexin V positive cells were increased in a dose-dependent manner. In addition to RBL-2H3 cells, the IL-4 emission from the other two cell lines was very low, and the TNF-a quantity detected by the three cell lines was also very small. The increase of the TNF-a positive rate of the three cell lines was significantly higher than that of the other three indexes. The 尾 -glucosaminidase release rate and Annexin V positive cell rate of P815 and Ku812 cells were increased earlier than that of RBL-2H3 cells under the same stimulation conditions.
