【摘要】：目的基于羥丙甲纖維素(HPMC)的壓縮性質(zhì),研究HPMC粒徑對硝苯地平親水凝膠骨架緩釋片釋放度影響的機(jī)制。方法測定不同粒徑HPMC K4M的壓縮性質(zhì),以硝苯地平為模型藥物,HPMC K4M為骨架材料,制備親水凝膠骨架緩釋片。研究HPMC K4M的壓縮性質(zhì)對緩釋片孔隙率、釋放度等性質(zhì)的影響。結(jié)果隨著粒徑的減小,HPMC K4M的堆密度、振實(shí)密度、壓縮度、抗張強(qiáng)度逐漸增大,彈性恢復(fù)逐漸減小,使得硝苯地平緩釋片的厚度和孔隙率逐漸減小,單位體積HPMC濃度逐漸增大,導(dǎo)致HPMC凝膠速率加快,使得水分滲入骨架片速率減慢,骨架膨脹減慢,釋放度減小。結(jié)論不同粒徑分布HPMC的壓縮性質(zhì)明顯不同,從而顯著影響硝苯地平緩釋片的孔隙率和凝膠骨架的形成速率,進(jìn)而影響緩釋片的釋放度。
[Abstract]:Objective to study the mechanism of the effect of HPMC particle size on the release of nifedipine hydrophilic gel matrix sustained-release tablets based on the compression properties of HPMC. Methods the compression properties of HPMC K4M with different diameters were determined. The hydrophilic gel matrix sustained-release tablets were prepared with nifedipine as the model drug HPMC K4M. The effects of compression properties of HPMC K4M on the porosity and release of HPMC K4M were studied. Results the bulk density of HPMC K4M decreased with the particle size of HPMC K4M. The density, compression, tensile strength and elastic recovery of nifedipine sustained-release tablets gradually increased, and the thickness and porosity of nifedipine sustained release tablets gradually decreased, and the HPMC concentration per unit volume gradually increased, which led to the acceleration of HPMC gel rate. The results showed that the rate of water permeation into the matrix decreased, the expansion of the matrix decreased and the release rate decreased. Conclusion the compression properties of HPMC with different particle size distributions are obviously different, which has a significant effect on the porosity of nifedipine sustained-release tablets and the formation rate of gel skeleton. Furthermore, the release of sustained release tablets was affected.
