【摘要】：目的:建立測定復方制劑中鹽酸賴氨酸含量的方法。方法:采用鄰苯二甲醛(OPA)柱前衍生化高效液相色譜法。色譜柱為Kromasil C18,流動相為0.04 mol/L磷酸二氫鈉溶液(10%氫氧化鈉溶液調(diào)節(jié)p H至7.2)-甲醇-乙腈(70∶15∶15,V/V/V),流速為1.5 ml/min,檢測波長為338 nm,柱溫為35℃,供試品與OPA衍生劑及硼酸鹽緩沖液按設定程序自動混勻后進樣。結(jié)果:鹽酸賴氨酸質(zhì)量濃度在0.027 4~0.438 8 mg/ml范圍內(nèi)與峰面積呈良好的線性關系(r=0.999 5);精密度、穩(wěn)定性、重復性試驗的RSD≤0.84%;在五維賴氨酸顆粒、賴氨肌醇維B12口服溶液、賴氨酸磷酸氫鈣片中鹽酸賴氨酸的平均加樣回收率分別為99.8%(RSD=0.86%,n=9)、100.1%(RSD=0.79%,n=9)、99.6%(RSD=0.65%,n=9)。對3批含鹽酸賴氨酸的復方制劑進行含量測定,所得結(jié)果與原質(zhì)量標準測定方法中的結(jié)果一致。結(jié)論:該方法樣品處理簡單快速,可用于含有鹽酸賴氨酸的復方制劑的質(zhì)量控制。
