【摘要】：目的:建立一種有效、靈敏、準(zhǔn)確的可用于檢測(cè)人腦脊液內(nèi)萬(wàn)古霉素濃度的方法學(xué)。方法:色譜柱:Diamonsil C18(200 mm×4.6 mm,5μm),流動(dòng)相:乙腈-0.05 mol·L-1磷酸二氫鈉溶液(pH3.6)(9∶91),流速:0.8 mL·min-1,紫外檢測(cè)波長(zhǎng):236 nm,內(nèi)標(biāo):去甲萬(wàn)古霉素,柱溫:40℃,進(jìn)樣量:20μL。結(jié)果:萬(wàn)古霉素峰與其他藥物峰分離良好,不受腦脊液內(nèi)源性物質(zhì)干擾。在0.40~101.00μg·mL-1范圍內(nèi)萬(wàn)古霉素濃度和兩者峰面積比值呈線性關(guān)系(r=0.999 6),最低定量限0.40μg·mL-1。萬(wàn)古霉素和內(nèi)標(biāo)的保留時(shí)間分別為13 min和10 min,日內(nèi)和日間RSD均小于10%,低、中、高3個(gè)濃度提取回收率分別為60%、30.6%、28.8%。結(jié)論:此方法具有較高可行性,準(zhǔn)確度高,適用于腦脊液中萬(wàn)古霉素濃度檢測(cè)。
