基于高分辨質譜技術的整體動物體內藥物成像分析新方法研究

【摘要】:質譜分子成像技術作為體內藥物分析的新興工具受到越來越多的關注,其中生物體內復雜基質的干擾是面臨的關鍵問題之一。本研究利用高分辨質譜技術的優(yōu)勢,采用空氣動力輔助離子化質譜成像方法(AFAI-MSI),建立了高特異性的整體動物體內藥物成像分析新方法,以提高體內藥物成像分析結果的準確性和可靠性。以候選新藥右旋娃兒藤寧堿(S-(+)-deoxytylophorinidine,CAT)為研究對象,采用高分辨質譜全掃描與靶向掃描相結合的方式,對整體動物體內藥物的分布進行AFAI-MSI分析。結果表明,該方法能夠準確地反映藥物特異性分布和處置情況。通過一次質譜成像實驗,從整體動物水平同時實現了候選新藥體內分布和藥物干預下內源性代謝物的成像分析,為候選新藥的早期研發(fā)提供思路與手段。
[Abstract]:As a new tool of drug analysis in vivo, mass spectrometry molecular imaging technology has attracted more and more attention. The interference of complex matrix in biological body is one of the key problems. In this study, the advantage of high resolution mass spectrometry (HRMS) is used to use aerodynamic assisted ionization mass spectrometry imaging (AFAI-MSI). A new method for the analysis of in vivo drug imaging of whole animal was established. In order to improve the accuracy and reliability of in vivo drug imaging analysis, a new drug, dextroverine, S-deoxytylophorinidine, was used. High resolution mass spectrometry (HRMS) combined with targeted scanning was used to analyze the distribution of drugs in whole animal body by AFAI-MSI. This method can accurately reflect the specific distribution and disposal of drugs. The imaging analysis of endogenous metabolites under drug intervention and distribution in vivo were realized simultaneously from the whole animal level, which provided ideas and means for the early research and development of new drug candidates.
