麥胚凝集素修飾的EGCG-明膠-殼聚糖納米粒的制備、表征及體外抗腫瘤活性研究

【摘要】:目的制備麥胚凝集素(WGA)修飾的表沒食子兒茶素沒食子酸酯(EGCG)-明膠(Gel)-殼聚糖(Cs)納米粒,研究其對結(jié)腸癌HT-29細(xì)胞的作用。方法以明膠及殼聚糖為載體輔料,采用靜電自組裝的方法制備EGCG-Gel-Cs納米粒,以包封率為指標(biāo),通過Box-Behnken設(shè)計(jì)-效應(yīng)面法(BBD-RSM)優(yōu)化納米粒的制備處方及工藝,再將經(jīng)戊二醛活化的WGA修飾到納米粒表面。采用差示掃描量熱分析藥物在納米粒中的存在狀態(tài),采用細(xì)胞毒性及細(xì)胞凋亡實(shí)驗(yàn),比較研究納米粒與原料藥的體外抗腫瘤活性。結(jié)果 EGCG-Gel-Cs納米粒的最優(yōu)處方:Gel與Cs的質(zhì)量比為4.2,Gel與EGCG的質(zhì)量比為2.82,反應(yīng)溫度為34℃,包封率為(74.42±0.074)%。EGCG-Gel-Cs納米粒粒徑為(264.13±6.48)nm,經(jīng)過修飾后納米粒粒徑為(389.70±9.00)nm。WGA-EGCG-Gel-Cs納米粒對HT-29細(xì)胞的細(xì)胞毒性和細(xì)胞凋亡率顯著高于EGCG原料藥。結(jié)論 WGA-EGCG-Gel-Cs納米?娠@著提高細(xì)胞毒性作用。
[Abstract]:Objective to prepare the epigallocatechin gallate (EGCG) -gelatin Gelin-chitosan (CS) nanoparticles modified by wheat germ agglutinin (WGA). Methods EGCG-Gel-Cs nanoparticles were prepared by electrostatic self-assembly with gelatin and chitosan as the carrier excipients. The encapsulation efficiency was taken as the index. The preparation of nanoparticles was optimized by Box-Behnken design-effect surface method (BBD-RSM). Then the WGA activated by glutaraldehyde was modified to the surface of nanoparticles. The presence of drugs in nanoparticles was analyzed by differential scanning calorimetry (DSC). Cytotoxicity and apoptosis were tested. The in vitro antitumor activity of EGCG-Gel-Cs nanoparticles was compared with that of raw materials. Results the mass ratio of EGCG-Gel-Cs nanoparticles to CS was 4. 2.
