甲磺酸多黏菌素對大鼠巨噬細(xì)胞免疫功能的影響

【摘要】:目的探究甲磺酸多黏菌素E(CMS)對大鼠腹腔巨噬細(xì)胞免疫功能的影響。方法用噻唑藍(lán)(MTT)法檢測大鼠腹腔巨噬細(xì)胞在細(xì)胞懸液(陰性對照組)和25,50,100,125,250,500μg·m L~(-1)CMS作用24 h后的細(xì)胞存活率。用酶聯(lián)免疫法測定50,125μg·m L~(-1)CMS作用3,6,12,24 h后,巨噬細(xì)胞腫瘤壞死因子-α(TNF-α)的分泌水平。用免疫熒光微球法測定50,125μg·m L~(-1)CMS作用3,6,12 h后,巨噬細(xì)胞的吞噬功能。結(jié)果與陰性對照組100.00%相比,25,50,100,250,500μg·m L~(-1)CMS作用后的細(xì)胞存活率分別為118.46%,124.72%,122.54%,46.64%,13.06%,差異均有統(tǒng)計(jì)學(xué)意義(P0.01);125μg·m L~(-1)CMS作用后的細(xì)胞存活率為102.60%,差異無統(tǒng)計(jì)學(xué)意義(P0.05)。作用3 h后,陰性對照組與125μg·m L~(-1)CMS給藥組的TNF-α水平分別為32.51 pg·m L~(-1),56.09 pg·m L~(-1),差異有統(tǒng)計(jì)學(xué)意義(P0.05)。作用6 h后,陰性對照組與50,125μg·m L~(-1)CMS給藥組的吞噬百分率分別為70.93%,82.03%,80.80%,陰性對照組與50,125μg·m L~(-1)CMS給藥組比較差異均有統(tǒng)計(jì)學(xué)意義(P0.01)。結(jié)論 CMS能通過提高細(xì)胞活性、促進(jìn)細(xì)胞因子分泌及增強(qiáng)細(xì)胞吞噬功能等途徑增強(qiáng)巨噬細(xì)胞的免疫功能,進(jìn)而發(fā)揮其免疫調(diào)節(jié)作用。
[Abstract]:Objective to investigate the effect of polymyxine mesylate (PMS) on the immune function of peritoneal macrophages in rats. And 25. The cell survival rate of 50,100,125,250,500 渭 g 路mL ~ (-1) CMS for 24 h was determined by enzyme-linked immunosorbent assay (Elisa). The levels of tumor necrosis factor- 偽 (TNF- 偽) in macrophages were determined by immunofluorescence microsphere method. The phagocytic function of macrophages was observed after 12 h treatment with 125 渭 g 路m ~ (-1) CMS. Results compared with the negative control group (100.00%), the phagocytic function of macrophages was 2550 / 100. The cell survival rate of 250,500 渭 g 路m ~ (-1) ~ (-1) CMS was 118.46% and 124.72% and 122.54 ~ 46.64%, respectively. 13.06, the difference was statistically significant (P 0.01); The cell survival rate was 102.60 after treatment with 125 渭 g 路mL ~ (-1) CMS, but there was no significant difference between the two groups (P 0.05). After 3 h of treatment, the cell survival rate was 102.60 渭 g 路mL ~ (-1). The levels of TNF- 偽 in the negative control group and 125 渭 g 路mL ~ (-1) CMS group were 32.51 PG 路m ~ (-1). 56.09 PG 路mL ~ (-1), the difference was statistically significant (P 0.05). After 6 hours of treatment, the negative control group was 50%. The percentage of phagocytosis in 125 渭 g 路mL ~ (-1) CMS group was 70.93 and 82.03, respectively, and that in negative control group was 80.80, and that in negative control group was 50. 125 渭 g 路mL ~ (-1) CMS could improve cell activity by increasing the cell activity, and the difference was statistically significant (P 0.01). Promoting cytokine secretion and enhancing phagocytic function can enhance the immune function of macrophages and then exert its immunomodulatory effect.
