和厚樸酚與siRNA共遞送的陽離子脂質(zhì)體研究

【摘要】:目的利用聚乙二醇(PEG)修飾的陽離子脂質(zhì)體實現(xiàn)和厚樸酚與siRNA的共遞送以改善腫瘤治療效應(yīng)。方法利用薄膜分散法制備聚乙二醇修飾的陽離子脂質(zhì)體,通過疏水作用包載和厚樸酚以及靜電吸附作用包載siRNA的方式,構(gòu)建共遞送和厚樸酚與siRNA的陽離子脂質(zhì)體。通過粒徑、電位、包封率、血清穩(wěn)定性等方法考察脂質(zhì)體的制劑學(xué)性質(zhì),并考察了制劑的細胞攝取與胞內(nèi)分布、溶酶體逃逸能力以及腫瘤細胞生長抑制效應(yīng)。結(jié)果所制備的各組載和厚樸酚脂質(zhì)體的平均粒徑均在100 nm以內(nèi),均具有較高的藥物包封率。優(yōu)選的共遞送脂質(zhì)體制劑(N/P=5)具有較好的血液穩(wěn)定性,可獲得較高的細胞攝取和較好的胞內(nèi)溶酶體逃逸能力;和厚樸酚與siRNA的協(xié)同效應(yīng)具有較好的抑制腫瘤細胞生長效果。結(jié)論采用聚乙二醇修飾的陽離子脂質(zhì)體可以實現(xiàn)和厚樸酚和siRNA的共遞送并將可能有利于體內(nèi)腫瘤治療。
[Abstract]:Objective to utilize polyethylene glycol (PEG). The modified cationic liposome and the codelivery of honokiol and siRNA were used to improve the therapeutic effect of tumor. Methods Polyethylene glycol modified cationic liposomes were prepared by thin film dispersion. The cationic liposomes of codelivery and magnolol with siRNA were constructed by hydrophobic encapsulation and honokiol and electrostatic adsorption of siRNA. The entrapment efficiency was determined by particle size, potential and encapsulation efficiency. The pharmacological properties of liposomes were investigated by serum stability method. The cellular uptake and intracellular distribution of liposomes were also investigated. Results the average particle size of each group of liposomes and honokiol liposomes was less than 100 nm. The selected co-delivery liposome preparation (N / P ~ (5)) had better blood stability, higher cellular uptake and better intracellular lysosome escape ability. The synergistic effect of honokiol and siRNA can inhibit the growth of tumor cells. Conclusion the co-delivery of honokiol and siRNA can be realized by using cationic liposomes modified with polyethylene glycol. It is beneficial to the treatment of tumor in vivo.
