【摘要】：目的:采用全柱成像毛細(xì)管等電聚焦電泳法(WCID-CIEF),測定多肽樣品、胰島素、融合蛋白疫苗和HPV病毒樣顆粒的等電點(diǎn)。方法:采用50 mm×100μm石英涂層毛細(xì)管。聚焦方法設(shè)置:1.5 k V聚焦1 min,3 k V聚焦4 min/8 min;檢測波長:280 nm。結(jié)果:多肽樣品、激素樣品胰島素和融合蛋白疫苗的等電點(diǎn)分別為10.98、5.87和5.86;病毒樣顆粒樣品HPV 31L1、HPV 33L1、HPV 45L1、HPV 52L1和HPV 58L1的實(shí)測等電點(diǎn)分別為8.17、7.86、7.77、7.91和7.69,并且實(shí)測等電點(diǎn)與理論等電點(diǎn)基本一致。全柱成像毛細(xì)管等電聚焦法測定生物制品專屬性和精密度均符合驗(yàn)證要求。結(jié)論:該方法可用于生物制品的等電點(diǎn)分析。
[Abstract]:Objective: to determine the insulin of polypeptide samples by capillary isoelectric focusing electrophoresis with full column imaging. Isoelectric point of fusion protein vaccine and HPV virus-like particles. Methods: 50 mm 脳 100 渭 m quartz coated capillary was used. 3kV focusing 4 min/8; Results: the isoelectric point of insulin and fusion protein vaccine of peptide sample, hormone sample and fusion protein vaccine were 10.98 and 5.86, respectively. The measured isoelectric points of HPV 31L 1 HPV33L 1 HPV45L 1 HPV52L1 and HPV 58L1 were 8.17 ~ 7.86, respectively. 7.77-7.91 and 7.69. The measured isoelectric point is basically consistent with the theoretical isoelectric point. The specificity and precision of the whole column imaging capillary isoelectric focusing method for the determination of biological products meet the requirements of verification. This method can be used for isoelectric point analysis of biological products.
