【摘要】:目的:建立血漿中地塞米松的含量測定方法,并用此法探討平原組與急進高原組Wistar大鼠體內地塞米松的藥動學特征。方法:色譜柱固定相為Phenomene Gemini C18(4.6 mm×250 mm,5μm),流動相為甲醇-水(80∶20),流速為1.0ml·min-1,紫外檢測波長為240 nm,柱溫為25℃,內標為甲睪酮;7只Wistar大鼠于平原采血(平原組,P)后,相隔一周清洗期后急進高原(急進高原組,H),禁食12h后將0.056 g醋酸地塞米松片劑灌胃。于給藥前(0h,空白血漿)及給藥后0.67,1,1.5,2,3,4,6,8,12及24 h由眼眶后靜脈叢取血,采用高效液相色譜法測定Wistar大鼠單劑量灌胃后地塞米松血藥濃度。結果:血漿地塞米松濃度測定的線性范圍為80~1 600 ng·ml-1,r=0.999 2,最低檢測限為80 ng·ml-1。比較急進高原組與平原組大鼠體內地塞米松藥動學參數,發(fā)現其發(fā)生了顯著變化,藥時曲線下面積從(616.848±68.585)μg·L-1·h增大到(1 222.825±83.192)μg·L-1·h,峰濃度增大、半衰期縮短、體內平均駐留時間延長、總清除率降低,且差異均有統(tǒng)計學意義,P0.05。結論:急進高原后,地塞米松在Wistar大鼠體內代謝過程發(fā)生明顯變化,研究結果為平原和急進高原后臨床合理應用地塞米松提供參考依據。
