CD38在鼻咽癌側(cè)群細(xì)胞中高表達(dá)病通過(guò)影響ZAP70信號(hào)通路發(fā)揮其生物學(xué)效應(yīng)

[目的]通過(guò)對(duì)鼻咽癌側(cè)群細(xì)胞(SP細(xì)胞)的鑒定和生物學(xué)功能研究,探討CD38在鼻咽癌側(cè)群細(xì)胞中的作用及其可能機(jī)制。[方法]利用貝克曼庫(kù)爾特公司(Beckman Coulter)MoFloTM XDP型超高速細(xì)胞分選平臺(tái),鑒定、分選鼻咽癌細(xì)胞系SP細(xì)胞。在此基礎(chǔ)上,采用流式細(xì)胞術(shù)、microRNA芯片、實(shí)時(shí)定量PCR、Western Blotting等方法,分別從mRNA、microRNA和蛋白質(zhì)表達(dá)水平等不同層面,研究鼻咽癌SP細(xì)胞的生物學(xué)特性、CD38在鼻咽癌側(cè)群細(xì)胞中的表達(dá)水平及其可能作用機(jī)制。[結(jié)果]1.人鼻咽癌細(xì)胞系中SP細(xì)胞的鑒定及其生物學(xué)特性分析。利用SP細(xì)胞外排熒光活性染料Hoechst33342的特性,我們鑒定了4種鼻咽癌細(xì)胞系HK-1、C666-1、HNE1和HNE2中SP細(xì)胞的比例,結(jié)果發(fā)現(xiàn)HK-1和C666-1兩種細(xì)胞系中SP細(xì)胞比例較高,分別為5.2土0.72%和0.46±0.07%,而HNE1和HNE2細(xì)胞系中比例較低,分別為0.01%和0.02%。接下來(lái)我們選擇SP細(xì)胞比例較高的HK-1細(xì)胞進(jìn)行后續(xù)研究。SP細(xì)胞因其高表達(dá)ABC轉(zhuǎn)運(yùn)蛋白能夠外排Hoechst...

碩士學(xué)位論文^細(xì)胞增殖最為顯著(圖3-3 A)。同樣,平板克隆形成實(shí)驗(yàn)表明,分選得到的SP細(xì)胞和NSP細(xì)胞經(jīng)過(guò)10天的培養(yǎng),在相同條件下、相同時(shí)間內(nèi)SP細(xì)胞比NSP細(xì)胞形成較多克隆,克隆體積也較大。形成相同大小克隆時(shí)SP細(xì)胞所用時(shí)間較短。最終,SP細(xì)胞的克隆形成效率為(15.4±2.9) %,NSP細(xì)胞的克隆形成效率為(4.5±1.7) %,t=5.561, p=0.0051,兩者有統(tǒng)計(jì)學(xué)差異(圖3-3B)。腫瘤微球形成實(shí)驗(yàn)中,在選擇性培養(yǎng)條件下,與NSP細(xì)胞相比,SP細(xì)胞形成了更多以及更大的腫瘤微球,t=6.608,p< 0.0001,差異有統(tǒng)計(jì)學(xué)意義(圖3-3 C)。通過(guò)以上實(shí)驗(yàn)提示,SP細(xì)胞比NSP細(xì)胞有較強(qiáng)的生長(zhǎng)和自我更新能力,這些特性與腫瘤千細(xì)胞極其相似。
