文章摘要:目的觀察異種(鼠)脫細(xì)胞真皮基質(zhì)作為結(jié)膜生長支架移植材料修復(fù)兔結(jié)膜缺損的效果,探討異種脫細(xì)胞真皮基質(zhì)應(yīng)用于結(jié)膜缺損修復(fù)和結(jié)膜囊狹窄成形術(shù)的可行性。方法60只健康新西蘭大白兔隨機(jī)分為實驗組(n=30)和對照組(n=30)。實驗組和對照組分別移植異種(鼠)脫細(xì)胞真皮基質(zhì)植片和同種異體鞏膜植片。于術(shù)后1周、2周、4周、8周、12周采集實驗兔耳動脈血,離心后行酶聯(lián)免疫吸附法檢測血清中白細(xì)胞介素-2(interleukin2,IL-2)、白細(xì)胞介素-10(interleukin10,IL-10)含量,同時取移植區(qū)組織行HE染色,光鏡下觀察植片組織學(xué)變化。結(jié)果實驗組和對照組植片均存活,實驗組和對照組結(jié)膜缺損修復(fù)時間分別為(16.33±0.77)d、(15.78±0.88)d,兩組比較差異無統(tǒng)計學(xué)意義(P=0.056)。組織學(xué)檢查顯示兩組術(shù)后早期植片中細(xì)胞浸潤,術(shù)后2周異種脫細(xì)胞真皮基質(zhì)中新生血管長入,,術(shù)后12周異種脫細(xì)胞真皮基質(zhì)被重建,異體鞏膜被吸收。術(shù)后實驗組與對照組外周血清IL-2、IL-10濃度差異均無統(tǒng)計學(xué)意義(均為P〉0.05);實驗組外周血清IL-2、IL-10濃度差異也均無統(tǒng)計學(xué)意義(均為P〉0.05)。結(jié)論異種脫細(xì)胞真皮基質(zhì)可能成為一種理想的結(jié)膜缺損修復(fù)和結(jié)膜囊狹窄成形術(shù)移植材料。
Abstr:Objective To observe the effects of applying xenogeneic (rat) acel- lular dermal matrix(rat-ADM) for conjunctival defect repair of rabbit,and investigate the feasibility of xenogeneic aceUular dermal matrix(xeno-ADM) for the conjunctival defect repair or conjunctival sac reconstruction. Methods Sixty health New Zealand white rabbits were randomly divided into the experimental group (30 cases) and control group (30 cases). The rabbits in the experimental group and control group were transplanted with rat-ADM graft and the allograft scleral graft, respectively. The peripheral blood were collected in experimental and control groups at 1 week, 2 weeks, 4 weeks, 8 weeks, 12 weeks after the operation, IL-2 and IL-10 concentrations in serum were meas- ured after centrifugation. Meanwhile, the tissues of transplant district were harvested for HE staining,then histologic changes were observed under fight-microscopy. Results The graft in experimental group and control group were all surviving, the conjunctival defect repair time in experimental group and control group were ( 15.33 ± 0.77 ) days and ( 15.78 ± 0.88 ) days, respectively, there was no statistical difference (P = 0.055 ). His- tological examination showed cells infiltrated in postoperative grafts of two groups ear- ly,new blood vessels grew in ADM grafts at 2 weeks after operation, and ADM grafts were rebuilted and sclera grafts were absorbed gradually at 12 weeks after operation. There was no statistical difference in IL-2 and IL-10 concentrations between two groups (all P 〉 0.05 ), and no statistical difference between IL-2 and IL-10 concentration in ex- perimental group ( all P 〉 0.05 ).
