心肌梗死與心室重構(gòu)中miRNA和轉(zhuǎn)錄因子共調(diào)控網(wǎng)絡(luò)分析

心肌梗死是當今社會中引發(fā)死亡的重要因素,因此開發(fā)新的治療方法是臨床和研究心肌梗死的重要目標。心肌梗死涉及到的相關(guān)基因和過程眾多,其中的調(diào)控關(guān)系也很復雜。本文中,我們基于基因調(diào)控網(wǎng)絡(luò)的方法對心肌梗死中的基因調(diào)控模式進行了綜合分析。首先,我們從多個資源如:PubMed、CADgene、GWAS、HMDD and miR2disease收集了與心肌梗死相關(guān)的基因和miRNA。其次,我們預測了人類所有轉(zhuǎn)錄因子、心肌梗死相關(guān)的miRNA及基因間的調(diào)控關(guān)系。轉(zhuǎn)錄因子對心肌梗死相關(guān)的miRNA和基因的調(diào)控,我們從UCSC(hg18)獲得了預測的轉(zhuǎn)錄因子結(jié)合位點以及從TRANSFAC和ChIP-seq數(shù)據(jù)獲得了轉(zhuǎn)錄因子驗證的靶基因。通過整合兩個miRNA靶基因預測軟件TargetScan和miRanda的結(jié)果,得到了miRNA預測的靶基因,并整合miR2Disease、miRTarBase、miRecords和TarBase四個數(shù)據(jù)庫收集的實驗驗證的miRNA靶基因,我們獲得了心肌梗死相關(guān)miRNA的靶基因。最后,我們使用上述調(diào)控關(guān)系構(gòu)建了心肌梗死特異的miRNA-轉(zhuǎn)錄因子共調(diào)控網(wǎng)絡(luò)。該網(wǎng)絡(luò)共包含了2...
