骨化三醇對(duì)脂多糖誘導(dǎo)主動(dòng)脈瓣膜間質(zhì)細(xì)胞成骨分化的影響

背景:在鈣化性主動(dòng)脈瓣膜病的發(fā)病過(guò)程中,主動(dòng)脈瓣膜間質(zhì)細(xì)胞發(fā)生成骨分化可促進(jìn)瓣膜鈣化。人體內(nèi)維生素D3缺乏與心血管疾病的發(fā)生及進(jìn)展密切相關(guān)。維生素D3重要的活性代謝產(chǎn)物-骨化三醇[1α,25-二羥維生素D3,1,25(OH)2D3]具有調(diào)節(jié)細(xì)胞的增殖、分化等重要的生物學(xué)作用,但它是否能影響主動(dòng)脈瓣膜間質(zhì)細(xì)胞成骨分化尚不清楚。目的:探究骨化三醇[1α,25-dihydroxycholecalciferol,1,25(OH)2D3]對(duì)脂多糖(Lipopolysaccharide,LPS)誘導(dǎo)豬主動(dòng)脈瓣膜間質(zhì)細(xì)胞成骨分化的影響,為鈣化性主動(dòng)脈瓣膜病的預(yù)防提供理論依據(jù)。方法:6只雄性健康家豬(810月齡,體重100120 kg),處死后即刻于無(wú)菌條件下取主動(dòng)脈瓣葉。膠原酶連續(xù)消化法分離主動(dòng)脈瓣膜間質(zhì)細(xì)胞,觀察其形態(tài)特征,并用免疫熒光染色行表型鑒定。利用條件培養(yǎng)基誘導(dǎo)細(xì)胞成骨分化,建立體外主動(dòng)脈瓣膜間...
