BMP9聯(lián)合Wnt3a誘導(dǎo)C3H10T1/2細(xì)胞向心肌細(xì)胞樣細(xì)胞分化的研究

第一部分?jǐn)U增重組腺病毒GFP、Wnt3a、BMP9并分別轉(zhuǎn)染C3H10T1/2細(xì)胞目的:擴(kuò)增能高度表達(dá)GFP、Wnt3a、BMP9蛋白的重組腺病毒,并檢測(cè)分別轉(zhuǎn)染C3H10T1/2細(xì)胞后的穩(wěn)定性。方法:復(fù)蘇HEK293細(xì)胞并培養(yǎng),待其增殖生長(zhǎng)至約培養(yǎng)瓶底面積的70%~80%,進(jìn)行反復(fù)傳代兩次后,向細(xì)胞中分別加入適量的AdEasyGFP、AdEasyWnt3a、AdEasyBMP9原液,搖晃均勻后培養(yǎng)48-72h觀察細(xì)胞形態(tài)發(fā)生了明顯的改變后,即可收集細(xì)胞懸液。于倒置相差顯微鏡下觀察復(fù)蘇培養(yǎng)的C3H10T1/2細(xì)胞,待其生長(zhǎng)鋪滿至約培養(yǎng)瓶底面積的70%時(shí),加入適量已成功收集的腺病毒液,48h后利用熒光顯微鏡和流式細(xì)胞術(shù)分別觀察腺病毒轉(zhuǎn)染后的細(xì)胞生長(zhǎng)變化及轉(zhuǎn)染效率。結(jié)果:1、AdEasyGFP、AdEasyWnt3a、AdEasyBMP9在HEK293細(xì)胞中反復(fù)擴(kuò)增后,細(xì)胞形態(tài)發(fā)生明顯改變,即收集到了能夠高度表達(dá)GFP、Wnt3a、BMP9蛋白的腺病毒液。2、鏡下觀察經(jīng)BMP9、Wnt3a及二者聯(lián)合轉(zhuǎn)染C3H10T1/2細(xì)胞1周后,可見BMP9組、Wnt3a聯(lián)合BMP9組細(xì)胞呈現(xiàn)纖維樣生長(zhǎng)...
