組織蛋白酶S通過Wnt5a/SC35信號通路調(diào)控老齡小鼠缺血誘導的血管再生機制

目的:本研究探討老齡小鼠缺血性血管再生過程中CatS介導的Wnt5a/SC35負性調(diào)控機制,進一步闡明衰老削弱血管再生的相關(guān)分子機制,為防治老年心血管疾病提供實驗依據(jù)。方法:1、動物實驗實驗1):老齡小鼠(25-29g,雄性,>24個月齡,n=31)(由日本名古屋大學提供),分為CatS基因敲除組(CatS-/-,n=16)和野生組(CatS+/+,n=15)。腹腔內(nèi)注射戊巴比妥(60mg/kg)進行麻醉,脫毛并切開左下腹皮膚,結(jié)扎股動脈根部及其主要分支后切除股動脈段的方法制作小鼠下肢缺血模型。造模成功后在特定時間點收集血液,腓腸肌和骨髓樣本,開展以下實驗:(1)通過激光散斑血流成像(LSBFI)測定術(shù)前、術(shù)后即刻、4天、7天、14天的下肢血流,用同一時間點的缺血側(cè)與非缺血側(cè)的下肢血流灌注比值分析血流恢復情況;(2)應(yīng)用CD31和Mac-3抗體進行免疫染色,c-Kit和Ki67抗體進行免疫熒光,評價肌肉組織中的毛細血管形成,巨噬細胞浸潤和骨髓中的干細胞增殖。(3)應(yīng)用流式細胞術(shù)(FACS)檢測骨髓及外周血中的血管內(nèi)皮祖細胞(EPC),評價EPC的增生動員;(4)使用酶聯(lián)免疫吸附檢...
