經(jīng)導(dǎo)管射頻消融去腎交感神經(jīng)術(shù)對高血壓犬左室肥厚及VEGFR1-PKG1-Calcineurin通路的影響

目的:明確經(jīng)導(dǎo)管射頻消融去腎交感神經(jīng)術(shù)(RDN)對高血壓犬左室肥厚的影響。其次,觀察經(jīng)導(dǎo)管射頻消融腎交感神經(jīng)對高血壓犬心肌血管內(nèi)皮生長因子(VEGF)-血管內(nèi)皮生長因子受體-1(VEGFR-1)-cGMP依賴性蛋白激酶-1(PKG-1)-鈣調(diào)神經(jīng)磷酸(Calcineurin)的變化,探討經(jīng)導(dǎo)管射頻消融腎交感神經(jīng)對改善左室肥厚的可能機制。方法:將比格犬(n=28)分為高脂組(n=22)和對照組(n=6),高脂組予以高脂飲食飼養(yǎng)建立高血壓犬模型,對照組給予普食。12周后,將建模成功的高血壓犬(n=20)隨機分組為手術(shù)組和假手術(shù)組每組10只,實驗過程中手術(shù)組1只、假手術(shù)組2只死亡;最后納入統(tǒng)計分析的比格犬?dāng)?shù)量為手術(shù)組(n=9)和假手術(shù)組(n=8)。手術(shù)組給予射頻消融腎交感神經(jīng)術(shù),假手術(shù)僅予以腎動脈造影。于基線、術(shù)前、術(shù)后(1、2、4、6月)用無創(chuàng)血壓計測定血壓;同時于基線、術(shù)前、術(shù)后6月行心臟彩超計算心臟各個腔徑、左室重量指數(shù)及E峰/A峰比值。術(shù)后6月處死比格犬,采血、取心臟組織。采用HE染色和電鏡觀察心肌細(xì)胞形態(tài);CD31免疫組織化學(xué)方法染心肌組織中毛細(xì)血管計算毛細(xì)血管密度。Western...
