腹主動脈瘤組織中HIF-1α、MMP-2及MMP-9的表達(dá)及意義

【摘要】:目的探討低氧誘導(dǎo)因子(HIF)-1α、基質(zhì)金屬蛋白酶(MMP)-2及MMP-9在腹主動脈瘤(AAA)組織中的表達(dá)及意義。方法采用免疫組化法檢測68例AAA及20例正常腹主動脈組織中HIF-1α、MMP-2及MMP-9的表達(dá),并分析三者與臨床病理參數(shù)的關(guān)系。結(jié)果 HIF-1α表達(dá)于AAA中膜及外膜平滑肌細(xì)胞、巨噬細(xì)胞的細(xì)胞核中,MMP-2表達(dá)于AAA中膜及外膜平滑肌細(xì)胞的細(xì)胞質(zhì)中,MMP-9表達(dá)于AAA中膜及外膜平滑肌細(xì)胞、巨噬細(xì)胞的細(xì)胞質(zhì)中,三者在正常腹主動脈組織中均不表達(dá)。AAA中HIF-1α、MMP-2及MMP-9的陽性率顯著高于正常腹主動脈(P0.001)。AAA中動脈瘤直徑≥5.0 cm的患者HIF-1α、MMP-2及MMP-9的表達(dá)高于動脈瘤直徑5.0 cm者(P0.001);AAA中有發(fā)生心腦血管意外史患者HIF-1α、MMP-2及MMP-9的表達(dá)高于未發(fā)生者(P0.001)。AAA中HIF-1α、MMP-2及MMP-9與年齡、性別、有無吸煙史及高血壓無關(guān)。同時AAA中HIF-1α、MMP-2及MMP-9表達(dá)兩兩呈正相關(guān)。結(jié)論 HIF-1α在AAA中高表達(dá),并與MMP-2及MMP-9起協(xié)同作用,在AAA的發(fā)生發(fā)展中起重要作用。
[Abstract]:Objective to investigate the expression and significance of hypoxia inducer (HIF)-1 偽, matrix metalloprotease (MMP)-2 and MMP-9 in (AAA) of abdominal aortic aneurysm. Methods Immunohistochemical method was used to detect the expression of HIF-1 偽, MMP-2 and MMP-9 in 68 cases of AAA and 20 cases of normal abdominal aortic tissue, and the relationship between them and clinicopathological parameters was analyzed. Results HIF-1 偽 was expressed in AAA media and outer membrane smooth muscle cells, MMP-2 was expressed in the nucleus of macrophages, MMP-2 was expressed in the cytoplasm of AAA media and outer membrane smooth muscle cells, and MMP-9 was expressed in AAA media and outer membrane smooth muscle cells. In the cytoplasm of macrophages, none of them were expressed in normal abdominal aortic tissue. HIF-1 偽 in AAA, The positive rate of MMP-2 and MMP-9 was significantly higher than that of normal abdominal aorta (P0.001).
