芳香化酶(CYP19A1)與絕經(jīng)后女性冠心病的關(guān)聯(lián)性研究

【摘要】:目的:芳香化酶基因CYP19A1基因多態(tài)性與冠心病的相關(guān)性目前仍然不清楚,本研究的目的是探討CYP19A1基因單核苷酸多態(tài)性在漢族、維吾爾族中與冠心病和循環(huán)性激素水平及芳香化酶水平之間的關(guān)系。方法:本研究納入1064例漢族(614例男性,450例女性)和790例維吾爾族人(484例男性,306例女性),在從中篩選出498例絕經(jīng)后女性(265例漢族,233例維吾爾族)。選擇了4個(gè)多態(tài)性位點(diǎn)(rs2236722,rs2304463,rs4646,rs4275794)利用(iMLDR)技術(shù)進(jìn)行基因分型。利用放射免疫法檢測(cè)雌二醇和睪酮;利用ELISA試劑盒檢測(cè)人血中芳香化酶水平。結(jié)果:在維吾爾族人群中,發(fā)現(xiàn)rs2236722和rs4646基因分型和冠心病有顯著的關(guān)聯(lián)性(P≤0.03,P≤0.001),其中rs4646的顯性模型(CCvsCA+AA)和相加模型(CAvs.CC+AA)具有明顯關(guān)聯(lián)(全部P≤0.001)。同時(shí)也發(fā)現(xiàn)在絕經(jīng)后女性中rs4646的基因型與性激素和芳香化酶水平有顯著關(guān)聯(lián)性(P≤0.001),對(duì)循環(huán)性激素和方向化酶水平進(jìn)行邏輯回歸調(diào)整混雜因素后和冠心病仍然有顯著關(guān)聯(lián)(雌二醇:OR=0.777,95%CI:0.717-0.842,P=0.000;芳香化酶水平:OR=0.953,95%CI:0.944-0.963,P=0.000),利用一般線(xiàn)性模型分析rs4646和激素水平的交互分析發(fā)現(xiàn),rs4646與芳香化酶水平顯著相關(guān)(P=0.026)。結(jié)論:芳香化酶基因CYP19A1的rs2236722和rs4646與冠心病、性激素、芳香化酶水平具有顯著關(guān)聯(lián)性。
[Abstract]:Objective: the association between CYP19A1 gene polymorphism of aromatase gene and coronary heart disease (CHD) is still unclear. The purpose of this study was to investigate the single nucleotide polymorphism of CYP19A1 gene in Han nationality. Relationship between coronary heart disease, circulating sex hormone levels and aromatase levels in Uighur. Methods: 1064 cases of Han nationality (614 males, 450 females) and 790 Uighurs (484 males and 306 females) were selected from this study. Among them, 498 postmenopausal women (265Han and 233Uyghur) were selected. Four polymorphic loci (rs2236722,rs2304463,rs4646,rs4275794) were selected for genotyping by (iMLDR). Estradiol and testosterone were measured by radioimmunoassay and aromatase levels in human blood were detected by ELISA kit. Results: there was a significant correlation between rs2236722 and rs4646 genotyping and coronary heart disease (CHD) in Uygur population (P < 0.001).
