冠心病患者外周血單個(gè)核細(xì)胞EPC百分比及SDF-1、VEGF-2水平變化及意義

【摘要】:目的探討冠狀動(dòng)脈粥樣硬化性心臟病(簡(jiǎn)稱(chēng)冠心病)患者外周血單個(gè)核細(xì)胞內(nèi)皮祖細(xì)胞(EPC)百分比及血清基質(zhì)細(xì)胞衍生因子1(SDF-1)、血管內(nèi)皮生長(zhǎng)因子2(VEGF-2)水平變化及意義。方法選取冠心病患者91例(冠心病組),體檢健康者40例(對(duì)照組)。采用流式細(xì)胞術(shù)測(cè)定外周血單個(gè)核細(xì)胞EPC百分比,ELISA法檢測(cè)血清SDF-1、VEGF-2;分離冠心病患者單個(gè)核細(xì)胞,分別給予0、10、20、40、80、160 ng/m L CXC族趨化因子受體4(CXCR4,SDF-1受體)拮抗劑AMD3100共培養(yǎng)7天,觀(guān)察各濃度AMD3100對(duì)EPC百分比及SDF-1、VEGF-2水平的影響。結(jié)果冠心病組外周血單個(gè)核細(xì)胞EPC百分比及SDF-1、VEGF-2水平均低于對(duì)照組,其中Ⅱ型斑塊冠心病患者三項(xiàng)指標(biāo)均低于Ⅰ型和Ⅲ型患者,三支冠狀動(dòng)脈狹窄患者三項(xiàng)指標(biāo)均低于單支和雙支患者,冠狀動(dòng)脈側(cè)支循環(huán)2、3級(jí)冠心病患者三項(xiàng)指標(biāo)均高于0、1級(jí)患者,組間及組內(nèi)比較P均0.05。隨著AMD3100濃度的升高,單個(gè)核細(xì)胞EPC百分比及血清SDF-1、VEGF-2水平依次降低,各濃度間比較P均0.05。結(jié)論冠心病患者外周血單個(gè)核細(xì)胞EPC百分比及血清SDF-1、VEGF-2水平均降低,三者的水平變化與冠心病的發(fā)生、發(fā)展有關(guān);通過(guò)SDF-1動(dòng)員EPC、VEGF-2,修復(fù)受損血管和形成新生血管,有望成為臨床治療冠心病的新靶點(diǎn)。
[Abstract]:Objective to investigate the percentage of endothelial progenitor cells (EPC) in peripheral blood mononuclear cells (PBMC) and serum stromal cell derived factor-1 (SDF-1) in patients with coronary atherosclerotic heart disease (CHD). Changes and significance of vascular endothelial growth factor 2 (VEGF-2) level. Methods 91 patients with coronary heart disease (CHD group) and 40 healthy persons (control group) were selected. The percentage of EPC in peripheral blood mononuclear cells was measured by flow cytometry and serum SDF-1,VEGF-2; was detected by ELISA method. Mononuclear cells were isolated from patients with coronary heart disease and were co-cultured for 7 days with AMD3100, a chemokine receptor 4 (CXCR4,SDF-1 receptor) antagonist of CXC group. The effect of AMD3100 concentration on EPC percentage and SDF-1,VEGF-2 level was observed.
