【摘要】：目的探究二脒那秦(DIZE)對肺動脈高壓(PAH)模型大鼠的預防及治療作用。方法采用左肺葉切除聯(lián)合野百合堿(MCT)腹壁皮下注射建立PAH大鼠模型,100只成年雄性Wistar大鼠隨機分成空白對照組(A組)、DIZE對照組(B組)、PAH模型組(C組)、DIZE處理PAH模型組(D組)和(DIZE+C-16)處理PAH模型組(E組),利用活性熒光共振能量轉移(FRET)和酶聯(lián)免疫吸附試驗(ELISA)法測定血管緊張素轉化酶2(ACE2)、血清IL-6及IL-8水平,并測定平均肺動脈壓(m PAP)、右心室肥厚指數(shù)(RVHI),計算中膜厚度與肺動脈外徑的比例(WT)、內膜增生評分,并通過彈力纖維染色分析肺血管病變。結果 RVHI、ACE2酶活性、WT及內膜增生評分比較:與A組比較,C組、D組和E組差異有統(tǒng)計學意義(P0.05);D組和E組差異有統(tǒng)計學意義(P0.05)。五個組在各個指標整體分析中差異有統(tǒng)計學意義(P0.05)。結論二脒那秦可以提高ACE2酶活性,并有效降低m PAP、RVHI和WT,同時減輕肺動脈中膜肥厚,并抑制肺小動脈內膜增生。該研究結論為二脒那秦治療人類PAH提供實驗基礎。
[Abstract]:Objective to investigate the preventive and therapeutic effects of dimidine (DIZE) on pulmonary hypertension (PAH) rats. Methods PAH rat model was established by left lobectomy and subcutaneous injection of monocrotaline (MCT) into abdominal wall. 100 adult male Wistar rats were randomly divided into control group (group A,), DIZE control group) and group B (), PAH model group, group C). PAH model group (D group) and PAH model group (DIZE C-16) were treated with DIZE. Angiotensin converting enzyme 2 (ACE2) was measured by active fluorescence resonance energy transfer (FRET) and enzyme-linked immunosorbent assay (ELISA). The levels of serum IL-6 and IL-8 were measured, and the mean pulmonary artery pressure (m PAP),) right ventricular hypertrophy index (RVHI),) was measured to calculate the ratio of intimal hyperplasia score (WT),) to the ratio of medial thickness to pulmonary artery diameter. Pulmonary vascular lesions were analyzed by elastic fiber staining. Results the activity of RVHI,ACE2, WT and intimal hyperplasia score: compared with group A, group C, group D and group E had significant differences (P0.05) between); D group and E group (P0.05). There were significant differences among the five groups in the whole analysis of each index (P0.05). Conclusion Diamidinazin can increase the activity of ACE2, decrease m PAP,RVHI and WT, reduce pulmonary artery intimal hypertrophy and inhibit intimal hyperplasia of pulmonary arterioles. The results provide experimental basis for the treatment of human PAH with dimidine.
