【摘要】：目的檢測(cè)microRNA-9-3p(miR-9-3p)在2型糖尿病(T2DM)患者以及T2DM合并不穩(wěn)定型心絞(T2DM+UA)痛患者血漿中的表達(dá)水平,分析miR-9-3p對(duì)于T2DM以及T2DM+UA的診斷價(jià)值。方法選取2012年1月-2013年6月唐山市工人醫(yī)院就診的單純T2DM患者50例為T2DM組,T2DM+UA患者50例為T2DM+UA組,健康體檢人員30例為對(duì)照組。抽取患者外周靜脈血,提取microRAN(miRNA),應(yīng)用實(shí)時(shí)熒光定量聚合酶鏈反應(yīng)(q RT-PCR),以miR-423-5p為內(nèi)參,檢測(cè)3組血漿中miR-9-3p表達(dá)水平,應(yīng)用受試者工作曲線(ROC)統(tǒng)計(jì)分析其對(duì)于T2DM以及T2DM+UA的診斷價(jià)值。結(jié)果 3組血漿miR-9-3p表達(dá)水平比較,差異均有統(tǒng)計(jì)學(xué)意義(P0.05),T2DM組及T2DM+UA組均高于對(duì)照組,T2DM+UA組高于T2DM組;miR-9-3p對(duì)于T2DM的診斷價(jià)值A(chǔ)UC=0.723(95%CI:0.610,0.837,P0.05),miR-9-3p對(duì)于T2DM+UA的診斷價(jià)值A(chǔ)UC=0.972(95%CI:0.963,0.981,P0.05)。結(jié)論 miR-9-3p在T2DM以及T2DM+UA患者血漿表達(dá)水平升高,miR-9-3p有望作為T2DM以及T2DM+UA早期診斷的分子標(biāo)志物。
[Abstract]:Objective to detect the expression of microRNA-9-3p (miR-9-3p) in plasma of patients with type 2 diabetes mellitus (T2DM) and patients with T2DM complicated with unstable cardiac strangulation (T2DM UA), and to analyze the diagnostic value of miR-9-3p for T2DM and T2DM UA. Methods from January 2012 to June 2013, 50 patients with T2DM were selected as T2DM group, 50 patients with T2DM UA as T2DM UA group and 30 healthy persons as control group. The peripheral venous blood was extracted and microRAN (miRNA), was extracted by real-time fluorescence quantitative polymerase chain reaction (Q RT-PCR). MiR-423-5p was used as internal reference to detect the expression of miR-9-3p in plasma of three groups. The diagnostic value of (ROC) for T2DM and T2DM UA was analyzed. Results there were significant differences in plasma miR-9-3p expression among the three groups (P0.05). The expression of miR-9-3p in T2DM group and T2DM UA group was higher than that in control group, and that in T2DM UA group was higher than that in T2DM group. The diagnostic value of miR-9-3p for T2DM (95 CI: 0.610 / 0.837) and miR-9-3p for T2DM UA (95 CI: 0.963 / 0.981p0.05). Conclusion the expression of miR-9-3p in plasma of patients with T2DM and T2DM UA is increased. MiR-9-3p may be a molecular marker for early diagnosis of T2DM and T2DM UA.
