透明質酸對游離脂肪酸引起的人主動脈血管平滑肌細胞表達IL-6、IL-8和TNF-α的影響

【摘要】:目的 探討透明質酸(HA)對游離脂肪酸(FFA)引起的人主動脈血管平滑肌細胞(HA-VSMC)表達白細胞介素6(IL-6)、IL-8和腫瘤壞死因子α(TNF-α)的影響。方法 培養(yǎng)HA-VSMC,用不同濃度(0、100、200、400μmol/L)FFA和不同濃度FFA+高分子量HA(HMW-HA)或低分子量HA(LMW-HA)處理細胞24、48 h。噻唑藍法檢測FFA和FFA+HA對HA-VSMC存活率的影響;Ed U染色檢測細胞增殖活性;酶聯(lián)免疫吸附法檢測培養(yǎng)上清IL-6、IL-8和TNF-α含量;實時熒光定量PCR和Western blot檢測IL-6、IL-8和TNF-αmRNA及蛋白表達水平。結果(1)FFA在400μmol/L內呈濃度依賴性誘導HA-VSMC增殖;(2)HMW-HA、LMW-HA均對FFA誘導的HA-VSMC增殖有抑制作用,且HMW-HA抑制作用強于LMW-HA;(3)FFA顯著促進HA-VSMC IL-6、IL-8和TNF-α的分泌,且有明顯的時間和濃度依耐性;HA可以抑制上述作用;(4)實時熒光定量PCR和Western blot結果顯示,FFA+HMWHA組、FFA+LMW-HA組IL-6、IL-8、TNF-αmRNA及蛋白表達水平均低于FFA組(均P0.01),其中FFA+HMW-HA組又低于FFA+LMW-HA組(P0.01)。結論 (1)FFA誘導HA-VSMC IL-6、IL-8、TNF-αmRNA和蛋白表達升高;(2)HA對FFA引起的HA-VSMC IL-6、IL-8和TNF-α的表達有抑制作用,且HMW-HA抑制效果強于LMW-HA。
[Abstract]:Objective to investigate the effect of hyaluronic acid (HA) on the expression of interleukin-6 (IL-6) IL-8 and tumor necrosis factor- 偽 (TNF- 偽) in human aortic vascular smooth muscle cells (HA-VSMC) induced by free fatty acid (FFA). Methods HA-VSMC was cultured and treated with different concentration (0100200400 渭 mol/L) FFA and FFA high molecular weight HA (HMW-HA) or low molecular weight HA (LMW-HA) for 24 h. The effect of FFA and FFA HA on the survival rate of HA-VSMC was detected by thiazolyl blue method. The proliferative activity was detected by Ed U staining, the IL-8 and TNF- 偽 contents in supernatants were detected by enzyme-linked immunosorbent assay (Elisa), the IL-8 and TNF- 偽 mRNA and protein expression levels were detected by real-time fluorescence quantitative PCR and Western blot. Results (1) FFA induced the proliferation of HA-VSMC in a concentration-dependent manner, (2) HMW-HALMW-HA inhibited the proliferation of HA-VSMC induced by FFA, and the inhibitory effect of HMW-HA was stronger than that of LMW-HA.The (3) FFA significantly promoted the secretion of IL-8 and TNF- 偽 in HA-VSMC IL-6. The results of real-time fluorescence quantitative PCR and Western blot showed that the expression levels of IL-6 and IL-8 TNF- 偽 mRNA and protein in FFA LMW-HA group were lower than those in FFA group (P0.01), and FFA HMW-HA group was lower than FFA LMW-HA group (P0.01). Conclusion (1) FFA induces an increase in the expression of TNF- 偽 mRNA and protein in HA-VSMC IL-6 and IL-8, and (2) HA can inhibit the expression of IL-8 and TNF- 偽 in HA-VSMC induced by FFA, and the inhibitory effect of HMW-HA is stronger than that of LMW-HA.
