熒光探針包被法測(cè)定血漿硫化氫濃度(英文)

【摘要】:目的:在熒光探針的基礎(chǔ)上建立一種簡(jiǎn)易的方法來(lái)檢測(cè)血液中的硫化氫(hydrogen sulfide,H2S)。方法:將熒光探針包被到96孔板上,冷藏待用。用飽和硫酸銨沉淀血漿或血清中的蛋白,離心后所得的上清液分別加到含包被探針和不含包被探針的微孔中,37℃避光孵育2 h,然后使用分光光度計(jì)(波長(zhǎng)λEx/λEm340/445 nm)讀取微孔的熒光值,計(jì)算包被有探針和相應(yīng)無(wú)探針微孔的熒光差值,根據(jù)標(biāo)準(zhǔn)曲線濃度計(jì)算出血液中H2S的濃度。結(jié)果:靈敏度和特異性測(cè)試表明,此方法檢測(cè)靈敏度下限可以達(dá)到0.3μmol/L,血液中其他成分甚至其他含硫活性成分和含硫氨基酸等對(duì)此方法影響甚微。應(yīng)用此方法檢測(cè)188名健康成年志愿者的血清H2S濃度[(12.1±3.5)μmol/L,95%CI:4.6~19.8μmol/L],所得結(jié)果呈正態(tài)分布(單樣本K-S檢驗(yàn),P0.1)。對(duì)30名高血壓患者和22名相匹配的健康志愿者血清H2S濃度檢測(cè)表明,前者H2S濃度低于后者[(3.52±1.49)μmol/L vs.(10.23±2.76)μmol/L],差異具有統(tǒng)計(jì)學(xué)意義(配對(duì)樣本t檢驗(yàn),t=9.937,P0.001)。檢測(cè)雄性Wistar大鼠血清[(19.66±2.32)μmol/L]和血漿[(18.67±2.07)μmol/L]以及動(dòng)脈血[(19.34±0.51)μmol/L]和靜脈血[(18.99±0.50)μmol/L]中的H2S濃度,結(jié)果表明雄性Wistar大鼠血清和血漿以及動(dòng)脈血和靜脈血兩者差異都沒(méi)有統(tǒng)計(jì)學(xué)意義(重復(fù)測(cè)量的方差分析,P=0.38)。樣品的重復(fù)性檢測(cè)穩(wěn)定性較好(兩因素方差分析,P0.05)。結(jié)論:此方法對(duì)檢測(cè)血液中的H2S具有簡(jiǎn)單、高靈敏度、特異性和可重復(fù)性等優(yōu)點(diǎn),出結(jié)果快,可適用于大樣本的高通量檢測(cè),滿足實(shí)驗(yàn)室基礎(chǔ)研究及臨床一次性大樣本H2S濃度檢測(cè)的需求。
