熒光探針包被法測定血漿硫化氫濃度(英文)

【摘要】:目的:在熒光探針的基礎上建立一種簡易的方法來檢測血液中的硫化氫(hydrogen sulfide,H2S)。方法:將熒光探針包被到96孔板上,冷藏待用。用飽和硫酸銨沉淀血漿或血清中的蛋白,離心后所得的上清液分別加到含包被探針和不含包被探針的微孔中,37℃避光孵育2 h,然后使用分光光度計(波長λEx/λEm340/445 nm)讀取微孔的熒光值,計算包被有探針和相應無探針微孔的熒光差值,根據標準曲線濃度計算出血液中H2S的濃度。結果:靈敏度和特異性測試表明,此方法檢測靈敏度下限可以達到0.3μmol/L,血液中其他成分甚至其他含硫活性成分和含硫氨基酸等對此方法影響甚微。應用此方法檢測188名健康成年志愿者的血清H2S濃度[(12.1±3.5)μmol/L,95%CI:4.6~19.8μmol/L],所得結果呈正態(tài)分布(單樣本K-S檢驗,P0.1)。對30名高血壓患者和22名相匹配的健康志愿者血清H2S濃度檢測表明,前者H2S濃度低于后者[(3.52±1.49)μmol/L vs.(10.23±2.76)μmol/L],差異具有統計學意義(配對樣本t檢驗,t=9.937,P0.001)。檢測雄性Wistar大鼠血清[(19.66±2.32)μmol/L]和血漿[(18.67±2.07)μmol/L]以及動脈血[(19.34±0.51)μmol/L]和靜脈血[(18.99±0.50)μmol/L]中的H2S濃度,結果表明雄性Wistar大鼠血清和血漿以及動脈血和靜脈血兩者差異都沒有統計學意義(重復測量的方差分析,P=0.38)。樣品的重復性檢測穩(wěn)定性較好(兩因素方差分析,P0.05)。結論:此方法對檢測血液中的H2S具有簡單、高靈敏度、特異性和可重復性等優(yōu)點,出結果快,可適用于大樣本的高通量檢測,滿足實驗室基礎研究及臨床一次性大樣本H2S濃度檢測的需求。
