【摘要】：目的建立流式細(xì)胞術(shù)檢測(cè)藥物依賴性血小板抗體的標(biāo)準(zhǔn)方法。方法針對(duì)1名再生障礙性貧血患者的血小板抗體檢測(cè),通過(guò)ELISA法檢測(cè)其血漿中和血小板上特異性和自身抗體,用Lifecodes LSATM Class I(Luminex試劑盒,磁珠液相懸浮芯片系統(tǒng))檢測(cè)患者血漿中HLA-Ⅰ類(lèi)抗體特異性;以流式細(xì)胞術(shù)檢測(cè)患者血漿中兩性霉素B依賴的血小板抗體。結(jié)果 ELISA法檢測(cè):血小板特異性抗體和自身抗體均為陰性;Luminex試劑盒測(cè)得:患者血清中HLA-Ⅰ類(lèi)抗體陽(yáng)性率為1%,未檢出有特異性的HLA-Ⅰ類(lèi)抗體。流式細(xì)胞術(shù)檢測(cè):患者血漿中存在兩性霉素B依賴的血小板抗體。結(jié)論流式細(xì)胞術(shù)或可成為檢測(cè)藥物依賴性血小板抗體的實(shí)驗(yàn)室標(biāo)準(zhǔn)手段,為臨床預(yù)防和治療藥物引起的血小板減少癥提供可靠的理論和技術(shù)保障。
[Abstract]:Objective to establish a standard method for detection of drug-dependent platelet antibodies by flow cytometry. Methods Platelet antibodies were detected by ELISA in a patient with aplastic anemia. The specific and autoantibodies in plasma and platelets were detected by Lifecodes LSATM Class I (Luminex kit. The specificity of HLA-I antibody in plasma was detected by magnetic bead suspension microarray system, and the platelet antibody dependent on amphotericin B in plasma was detected by flow cytometry. Results ELISA assay showed that both platelet specific antibody and autoantibody were negative Luminex test kit. The positive rate of HLA-I antibody in serum was 1 and no specific HLA-I antibody was detected. Flow cytometry: Amphotericin B-dependent platelet antibodies were present in patients' plasma. Conclusion flow cytometry may be a laboratory standard for the detection of drug-dependent platelet antibodies and provide a reliable theoretical and technical guarantee for clinical prevention and treatment of drug-induced thrombocytopenia.
