【摘要】:目的:探討攜帶血管內(nèi)皮生長因子165(vascular endothelial growth factor 165,VEGF165)基因的腺病毒(Ad-VEGF165)轉(zhuǎn)染至心肌樣細(xì)胞對其表達VEGF165及增殖活性的影響。方法:提取、培養(yǎng)、鑒定骨髓間充質(zhì)干細(xì)胞(bone marrow mesenchymal stem cells,BMSCs);體外誘導(dǎo)BMSCs為心肌樣細(xì)胞。構(gòu)建攜帶VEGF165基因的腺病毒載體并轉(zhuǎn)染至心肌樣細(xì)胞;實驗分為3組:Ad-VEGF165轉(zhuǎn)染心肌樣細(xì)胞作為實驗組;未攜帶VEGF165基因的腺病毒轉(zhuǎn)染心肌樣細(xì)胞作為空載體組;未處理的心肌樣細(xì)胞作為空白對照組。在1周內(nèi)檢測各組VEGF165的表達及心肌樣細(xì)胞的增殖活性和相關(guān)指標(biāo)表達。結(jié)果:VEGF165在實驗組可正常表達,與空載體組、對照組相比差異具有統(tǒng)計學(xué)意義(P0.05);心肌樣細(xì)胞增殖活性在實驗組中最佳,顯著優(yōu)于空載體組和對照組(P0.05)。結(jié)論:成功將Ad-VEGF165轉(zhuǎn)染至心肌樣細(xì)胞,且表達的VEGF165可增強心肌樣細(xì)胞的活性。
[Abstract]:Aim: to investigate the effect of adenovirus (Ad-VEGF165) carrying vascular endothelial growth factor 165 (VEGF 165) gene on the expression and proliferation of VEGF165 in cardiomyoid cells. Methods: bone marrow mesenchymal stem cells (bone marrow mesenchymal stem cells) were isolated, cultured and identified. The adenovirus vector carrying VEGF165 gene was constructed and transfected into cardiomyocyte-like cells. Untreated cardiomyoid cells served as blank control group. The expression of VEGF165, the proliferative activity of cardiomyocyte-like cells and the expression of related indexes were detected within 1 week. Results the expression of VEGF165 was normal in the experimental group, and the difference was statistically significant compared with the empty vector group and the control group (P0.05), and the proliferation activity of myocardial like cells was the best in the experimental group, which was significantly better than that in the empty vector group and the control group (P0.05). Conclusion: Ad-VEGF165 was successfully transfected into cardiomyocyte-like cells, and the expression of VEGF165 could enhance the activity of cardiomyocyte-like cells.
