過表達(dá)缺氧誘導(dǎo)因子1α心肌干細(xì)胞能提高心肌梗死后心功能及移植細(xì)胞的存活率

【摘要】:背景:干細(xì)胞移植后最大的障礙是細(xì)胞在缺血、缺氧等惡劣環(huán)境下極低的存活率。為解決這一難題,人們嘗試基因修飾的方法,即在細(xì)胞移植前將抗凋亡基因通過轉(zhuǎn)染或轉(zhuǎn)導(dǎo)的方式,使其基因結(jié)構(gòu)發(fā)生改變,進(jìn)而可能會(huì)從根源上解決干細(xì)胞的存活、代謝及增生或分化等能力。目的:經(jīng)缺氧誘導(dǎo)因子1α基因修飾的心肌干細(xì)胞移植入大鼠心肌梗死邊緣區(qū)后,觀察移植后心肌干細(xì)胞的存活率及大鼠心功能的變化。方法:將24只SD大鼠隨機(jī)等分為3組:缺氧誘導(dǎo)因子1α轉(zhuǎn)染心肌干細(xì)胞組、心肌干細(xì)胞組和模型組,以開胸結(jié)扎冠脈的方法制備心肌梗死模型,2周后分別注射經(jīng)Ad-HIF1α-eGFP轉(zhuǎn)染的心肌干細(xì)胞、Ad-eGFP轉(zhuǎn)染的心肌干細(xì)胞及PBS液。結(jié)果與結(jié)論:心肌干細(xì)胞移植可提高心功能;經(jīng)缺氧誘導(dǎo)因子1α基因修飾的心肌干細(xì)胞移植可較單純心肌干細(xì)胞移植更顯著改善心功能;梗死后左室射血分?jǐn)?shù)的改善與梗死邊緣區(qū)心肌干細(xì)胞密度呈正相關(guān)。說明過表達(dá)缺氧誘導(dǎo)因子1α的心肌干細(xì)胞能有效改善心肌梗死大鼠心功能及移植細(xì)胞的存活。
[Abstract]:Background: the biggest obstacle after stem cell transplantation is the very low survival rate of cells in poor conditions such as ischemia and hypoxia. In order to solve this problem, people try to modify the gene, that is, to change the gene structure of anti-apoptotic gene by transfection or transduction before cell transplantation, which may solve the root cause of stem cell survival. Metabolism and ability to proliferate or differentiate. Aim: to observe the survival rate and cardiac function of myocardial stem cells modified by hypoxia inducible factor 1 偽 gene after transplantation into the marginal area of myocardial infarction in rats. Methods: Twenty-four SD rats were randomly divided into three groups: hypoxia inducible factor-1 偽 transfected myocardial stem cells group, myocardial stem cell group and model group. Myocardial infarction model was established by opening and ligating the coronary artery. After 2 weeks, the myocardial stem cells transfected with Ad-HIF1 偽 -eGFP and PBS solution were injected with Ad-eGFP respectively. Results and conclusion: myocardial stem cell transplantation can improve cardiac function, and hypoxia inducible factor 1 偽 gene modified myocardial stem cell transplantation can improve cardiac function more significantly than myocardial stem cell transplantation alone. The improvement of left ventricular ejection fraction was positively correlated with myocardial stem cell density. These results suggest that myocardial stem cells overexpression of hypoxia inducible factor-1 偽 can effectively improve cardiac function and survival of transplanted cells in rats with myocardial infarction.
