牛蒡根水提物對高血壓血清致傷血管內皮細胞TLR4、NF-κB表達的影響

【摘要】:目的:探究牛蒡根水提物對高血壓患者血清致傷血管內皮細胞Toll樣受體-核轉錄因子(TLR-NF-κB)信號轉導通路的影響。方法:選取2014年12月至2016年1月于醫(yī)院診治的高血壓病患者40例納入高血壓組,選擇健康體檢者40例納入對照組。分別采用兩組研究對象的血清干預人臍靜脈內皮細胞(human umbilical vein endothelial cells,HUVEC-C)24h,應用不同濃度的牛蒡根水提取物干預高血壓患者血清誘導的HUVEC-C,Toll樣受體4(toll like receptor 4,TLR4)表達(24h)及核轉錄因子(nuclear factorκB,NF-κB)活化(2h)。將細胞連續(xù)培養(yǎng)24h后將細胞蛋白提取,并用免疫印跡(Western blot)技術檢測細胞內TLR4蛋白及NF-κB抑制因子I-κBα蛋白的表達。結果:(1)高血壓組細胞提取蛋白中的TLR-4水平為(0.6683±0.07468),明顯高于對照組的(0.2220±0.02749),I-κBα水平為(0.2687±0.02519),明顯低于對照組的(0.5616±0.07762),差異均有統(tǒng)計學意義(P0.05);(2)牛蒡根水提物干預低劑量組、中劑量組及高劑量組細胞蛋白中的TLR-4表達水平為(0.2720±0.05178)、(0.4509±0.03399)、(0.3810±0.07796),均低于高血壓組的(0.6683±0.07468),同時高于對照組的(0.2220±0.02749);I-κBα表達水平為(0.5059±0.03042)、(0.3433±0.02289)、(0.3653±0.00710),高于高血壓組的(0.2687±0.02519),同時低于對照組的(0.5616±0.07762),差異均有統(tǒng)計學意義(P0.05)。結論:經(jīng)TLR-NF-κB信號途徑介導的免疫紊亂和炎癥反應可能是高血壓病血管內皮損傷的機制之一,牛蒡根水提取物可通過抑制細胞TLR-4、NF-κB的表達,來抑制炎癥因子的表達,最后達到保護血管內皮細胞的作用。
[Abstract]:Aim: to investigate the effect of burdock root water extract on Toll-like receptor-nuclear transcription factor (TLR-NF- 魏 B) signal transduction pathway in serum injured vascular endothelial cells in patients with hypertension. Methods: 40 hypertensive patients treated in hospital from December 2014 to January 2016 were selected into the hypertension group and 40 healthy persons were selected as the control group. Human umbilical vein endothelial cells (human umbilical vein endothelial cells HUVEC-C) were treated with serum from two groups for 24 h. Different concentrations of Arctium lappa root water extract were used to interfere with the expression (24 h) and the activation of nuclear transcription factor (nuclear factor 魏 B) NF- 魏 B (2 h) of HUVEC-CT-Toll-like receptor 4 (toll like receptor 4 (TLR4) and the activation of nuclear transcription factor (nuclear factor 魏 B (NF- 魏 B) in hypertensive patients. After 24 hours of continuous culture, the cell protein was extracted, and the expression of TLR4 protein and NF- 魏 B inhibitor I- 魏 B 偽 protein was detected by Western blot. Results: (1) the level of TLR-4 in cell extract protein in hypertension group was (0.6683 鹵0.07468), significantly higher than that in control group (0.2220 鹵0.02749) and (0.2687 鹵0.02519), significantly lower than that in control group (0.5616 鹵0.07762).
