牛蒡根水提物對(duì)高血壓血清致傷血管內(nèi)皮細(xì)胞TLR4、NF-κB表達(dá)的影響

【摘要】:目的:探究牛蒡根水提物對(duì)高血壓患者血清致傷血管內(nèi)皮細(xì)胞Toll樣受體-核轉(zhuǎn)錄因子(TLR-NF-κB)信號(hào)轉(zhuǎn)導(dǎo)通路的影響。方法:選取2014年12月至2016年1月于醫(yī)院診治的高血壓病患者40例納入高血壓組,選擇健康體檢者40例納入對(duì)照組。分別采用兩組研究對(duì)象的血清干預(yù)人臍靜脈內(nèi)皮細(xì)胞(human umbilical vein endothelial cells,HUVEC-C)24h,應(yīng)用不同濃度的牛蒡根水提取物干預(yù)高血壓患者血清誘導(dǎo)的HUVEC-C,Toll樣受體4(toll like receptor 4,TLR4)表達(dá)(24h)及核轉(zhuǎn)錄因子(nuclear factorκB,NF-κB)活化(2h)。將細(xì)胞連續(xù)培養(yǎng)24h后將細(xì)胞蛋白提取,并用免疫印跡(Western blot)技術(shù)檢測(cè)細(xì)胞內(nèi)TLR4蛋白及NF-κB抑制因子I-κBα蛋白的表達(dá)。結(jié)果:(1)高血壓組細(xì)胞提取蛋白中的TLR-4水平為(0.6683±0.07468),明顯高于對(duì)照組的(0.2220±0.02749),I-κBα水平為(0.2687±0.02519),明顯低于對(duì)照組的(0.5616±0.07762),差異均有統(tǒng)計(jì)學(xué)意義(P0.05);(2)牛蒡根水提物干預(yù)低劑量組、中劑量組及高劑量組細(xì)胞蛋白中的TLR-4表達(dá)水平為(0.2720±0.05178)、(0.4509±0.03399)、(0.3810±0.07796),均低于高血壓組的(0.6683±0.07468),同時(shí)高于對(duì)照組的(0.2220±0.02749);I-κBα表達(dá)水平為(0.5059±0.03042)、(0.3433±0.02289)、(0.3653±0.00710),高于高血壓組的(0.2687±0.02519),同時(shí)低于對(duì)照組的(0.5616±0.07762),差異均有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:經(jīng)TLR-NF-κB信號(hào)途徑介導(dǎo)的免疫紊亂和炎癥反應(yīng)可能是高血壓病血管內(nèi)皮損傷的機(jī)制之一,牛蒡根水提取物可通過抑制細(xì)胞TLR-4、NF-κB的表達(dá),來抑制炎癥因子的表達(dá),最后達(dá)到保護(hù)血管內(nèi)皮細(xì)胞的作用。
[Abstract]:Aim: to investigate the effect of burdock root water extract on Toll-like receptor-nuclear transcription factor (TLR-NF- 魏 B) signal transduction pathway in serum injured vascular endothelial cells in patients with hypertension. Methods: 40 hypertensive patients treated in hospital from December 2014 to January 2016 were selected into the hypertension group and 40 healthy persons were selected as the control group. Human umbilical vein endothelial cells (human umbilical vein endothelial cells HUVEC-C) were treated with serum from two groups for 24 h. Different concentrations of Arctium lappa root water extract were used to interfere with the expression (24 h) and the activation of nuclear transcription factor (nuclear factor 魏 B) NF- 魏 B (2 h) of HUVEC-CT-Toll-like receptor 4 (toll like receptor 4 (TLR4) and the activation of nuclear transcription factor (nuclear factor 魏 B (NF- 魏 B) in hypertensive patients. After 24 hours of continuous culture, the cell protein was extracted, and the expression of TLR4 protein and NF- 魏 B inhibitor I- 魏 B 偽 protein was detected by Western blot. Results: (1) the level of TLR-4 in cell extract protein in hypertension group was (0.6683 鹵0.07468), significantly higher than that in control group (0.2220 鹵0.02749) and (0.2687 鹵0.02519), significantly lower than that in control group (0.5616 鹵0.07762).
