大豆提取物對家兔動脈粥樣硬化斑塊中MMP-9及TIMP-1表達的影響

【摘要】:目的:探討大豆提取物(SE)對家兔動脈粥樣硬化(As)斑塊中基質金屬蛋白酶-9(MMP-9)及金屬蛋白酶組織抑制劑-1(TIMP-l)表達的影響。方法:取本課題組前期動物實驗中制備的家兔頸動脈粥樣硬化血管組織蠟塊40件,其中,實驗對照組、As模型組及SE低、中、高劑量組每組各8件,采用免疫組化法檢測粥樣硬化斑塊中MMP-9和TIMP-l的表達。結果:與實驗對照組相比,As模型組MMP-9和TIMP-1表達量明顯升高,TIMP-1/MMP-9比值明顯降低(p0.05);而與As模型組相比,SE低、中、高劑量組MMP-9和TIMP-1表達量不同程度降低,TIMP-1/MMP-9比值不同程度升高(p0.05)。經(jīng)Pearson相關性分析,SE劑量與家兔頸動脈粥樣硬化斑塊中MMP-9和TIMP-1表達量之間呈負相關,相關系數(shù)(r)分別等于-0.720、-0.831(p0.05),與TIMP-1/MMP-9比值之間呈正相關,相關系數(shù)(r)等于0.870(p0.05)。結論:SE可促使高同型半胱氨酸血癥(HHcy)誘導的As家兔頸動脈粥樣硬化斑塊中MMP-9和TIMP-1的表達下調(diào),引起TIMP-1/MMP-9比值升高;SE劑量與HHcy誘導的動脈粥樣硬化家兔頸動脈粥樣硬化斑塊中MMP-9和TIMP-1的表達量以及TIMP-1/MMP-9比值之間存在一定的劑量效應關系。
[Abstract]:Aim: to investigate the effects of soybean extract (SEE) on the expression of matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP-l) in atherosclerotic plaque of rabbits. Methods: forty rabbit carotid atherosclerotic tissue wax blocks were prepared in the previous animal experiment of our group. Among them, the experimental control group had 8 wax lumps in each group with low as model and low SE, middle and high dose groups, and the control group was divided into two groups: the control group, the middle dose group, and the high dose group. The expression of MMP-9 and TIMP-1 in atherosclerotic plaques was detected by immunohistochemical method. Results: compared with the experimental control group, the expression of MMP-9 and TIMP-1 in the as model group was significantly increased and the ratio of TIMP-1 / MMP-9 was significantly decreased compared with that in the as model group, while the expression of MMP-9 and TIMP-1 in the high dose group was lower than that in the as model group, and the ratio of MMP-9 and TIMP-1 / MMP-9 was increased to different degrees in the high dose group. By Pearson correlation analysis, there was a negative correlation between the dose of SE and the expression of MMP-9 and TIMP-1 in carotid atherosclerotic plaques in rabbits. The correlation coefficient was r = -0.720 ~ (-1) -0.831% p0.05, respectively, and positively correlated with the ratio of TIMP-1 / MMP-9, the correlation coefficient (r) was 0.870 ~ (-1) p _ (0.05). Conclusion the expression of MMP-9 and TIMP-1 in atherosclerotic plaques of as rabbits induced by hyperhomocysteinemia can be down-regulated by% SE, and the expression of MMP-9 and TIMP-1 in atherosclerotic plaques of as rabbits can be down-regulated. There was a dose-effect relationship between the increase of TIMP-1 / MMP-9 ratio and the expression of MMP-9 and TIMP-1 in atherosclerotic plaque induced by HHcy and the ratio of TIMP-1 / MMP-9.
