動(dòng)脈粥樣硬化中Toll樣受體3上調(diào)β型干擾素的表達(dá)

【摘要】:目的觀察動(dòng)脈粥樣硬化病變中Toll樣受體3(TLR3)與β型干擾素(IFN-β)表達(dá)的關(guān)系及影響。方法下肢動(dòng)脈硬化性閉塞癥(ASO)患者20例,健康對照組20例,通過流式細(xì)胞術(shù)檢測外周血單核細(xì)胞TLR3及IFN-β的表達(dá)。高脂飲食喂養(yǎng)Apo E(-/-)小鼠,實(shí)驗(yàn)組使用聚肌胞苷酸Poly(I:C)腹腔注射,對照組注射等劑量生理鹽水。兩日注射一次,經(jīng)10周后,生化分析儀檢測血漿中低密度脂蛋白膽固醇、高密度脂蛋白膽固醇、三酰甘油、總膽固醇、葡萄糖(Glu)水平,ELISA法檢測血漿及動(dòng)脈中TLR3、干擾素調(diào)節(jié)因子3(IRF3)、IFN-β及白介素-1β(IL-1β)水平。結(jié)果下肢ASO患者外周血單核細(xì)胞TLR3及IFN-β在ASO組的表達(dá)陽性率均顯著高于對照組;Apo E(-/-)小鼠中,實(shí)驗(yàn)組血漿中Glu水平較對照組顯著降低;實(shí)驗(yàn)組血漿及動(dòng)脈中TLR3、IRF3、IFN-β水平較對照組顯著增加,IL-1β水平較對照組顯著降低。結(jié)論動(dòng)脈粥樣硬化病變中TLR3表達(dá)的增加誘導(dǎo)IFN-β,并通過下調(diào)IL-1β水平達(dá)到抗動(dòng)脈粥樣硬化的作用。
[Abstract]:Objective To observe the relationship between the expression of Toll like receptor 3 (TLR3) and interferon beta (IFN- beta) in atherosclerotic lesions. Methods 20 patients with lower limb arteriosclerotic occlusion (ASO) and 20 healthy controls were used to detect the expression of TLR3 and IFN- beta in peripheral blood mononuclear cells by flow cytometry. High fat diet fed Apo E (- / -) mice, experimental group. By intraperitoneal injection of polycytidine Poly (I:C) and injection of normal saline in the control group. After two days of injection, the plasma middle low density lipoprotein cholesterol, high density lipoprotein cholesterol, three acylglycerol, total cholesterol, glucose (Glu) level, TLR3 in plasma and artery were detected by ELISA, and interferon regulatory factors were detected by ELISA method. Results the positive rates of the expression of TLR3 and IFN- beta in peripheral blood mononuclear cells of lower extremity ASO patients were significantly higher than those in the control group. In the Apo E (/ -) mice, the Glu level in the experimental group was significantly lower than that of the control group, and the plasma and arterial TLR3 in the experimental group were significantly increased in the experimental group than in the control group, and the level of IL-1 beta and -1 beta (IL-1 beta) in the lower extremities ASO patients were significantly higher than that of the control group. Addition, IL-1 beta level was significantly lower than that in the control group. Conclusion the increase of TLR3 expression in atherosclerotic lesions induced IFN- beta and the anti atherosclerotic effect by lowering the level of IL-1 beta.
